摘要
以多叶型苜蓿鲜草WL343HQ为原料,用酸化加热法提取苜蓿叶蛋白。研究提取温度、提取液p H和提取时间三因素对多叶型苜蓿叶蛋白提取率的影响。在单因素试验基础上,采用三因素三水平正交试验确定多叶型苜蓿叶蛋白提取的最佳工艺条件为:提取温度为50℃,p H为3,时间为30 min,在此工艺条件下,粗蛋白提取率为40.79%。通过氨基酸分析结果表明,多叶型苜蓿叶蛋白氨基酸种类齐全,含量高,氨基酸比例协调,因此多叶型苜蓿叶蛋白是优质蛋白质。
With multifoliolate alfalfa WL343 HQ as raw material, alfalfa leaf protein was extracted with acidification heating method. The effects of three factors such as extraction temperature, p H and extraction time on extraction rate of multifoliolate alfalfa leaf protein was discussed. On the basis of single factor experiments, the optimal conditions for hot acid extraction were achieved by three factors and three levels of orthogonal experiment, namely extraction temperature 50 ℃, p H 3 and the extraction time 30 min. Under the technical conditions, the extraction rate of crude protein was 40.79%. Amino acids analysis was employed, and the results show that the type of amino acids in leaf protein from multifoliolate alfalfa was complete, high content, and the proportion coordination of amino acids. Therefore, leaf protein from multifoliolate alfalfa was a kind of high quality protein.
