摘要 目的观察PKH26标记人脐带间充质干细胞(umbilical cord mesencymal stem cells,UC-MSCs)在大鼠子宫内膜的迁移分布情况,评价PKH26的标记和示踪效果。方法分离、培养UC-MSCs,应用PKH26进行细胞标记,将标记细胞经尾静脉移植入大鼠体内,荧光显微镜下观察细胞向子宫内膜迁移情况。结果 PKH26标记后细胞形态和生长活力无明显差别,荧光显微镜下可见几乎所有标记细胞呈红色荧光,主要分布于大鼠子宫内膜腺上皮和间质中。结论 PKH26荧光标记是一种安全有效的标记UC-MSCs的方法,可以用于干细胞移植方面的研究。 Objective By labeling the human umbilical cord mesenchymal stem cells(UCMSCs)with PKH26,the migration of cell in endometrium of rats were observed.Labeling and tracing effects of PKH26 were evaluated.Methods Isolated human UC-MSCs were labeled with PKH26.The labeled cells were transplanted to rats through caudal vein.Then the distribution of cells in rat endometrium was observed under fluorescence microscope.Results There were no detectable differences in cell morphology and growth after PKH26 labeling.And the cells were mainly labeled with red fluorescence under inverted fluorescence microscope and were mainly distributed in epithelial cells and stromal cells,in endometrial glandular epithelium and mesenchyme.Conclusion Labeling the human UC-MSCs with PKH26 is a safe and effective method,which can be used in the study of transplantation of stem cells.
