摘要 【目的】探索VEGF及其受体(VEGFR2)在繁殖期和繁殖间期成年牦牛睾丸组织中的分布特点并进行比较分析.【方法】应用免疫组织化学SP法检测VEGF及其受体(VEGFR2)分布特征并通过IPP图像分析软件进行定量统计.【结果】免疫组织化学显示,成年牦牛睾丸中,VEGF表达于各级生精细胞以及Sertoli细胞和Leydig细胞,强表达于长形精子,小血管内皮细胞见强阳性表达,肌样细胞未见表达;其受体VEGFR2表达于各级生精细胞及Sertoli细胞,且Leydig细胞内强表达,小血管弱表达;性成熟牦牛睾丸组织中VEGF和VEGFR2的表达繁殖期高于繁殖间期,但无显著统计学差异(P>0.05).【结论】VEGF及其受体在性成熟牦牛睾丸组织中表达丰富,表明其参与精子发生及局部微环境的调节,不同繁殖期有一定的表达差异,提示其表达与牦牛发情活动密切相关. 【Objective】To explore and analyze the distribution characteristics of VEGF and its receptor(VEGFR2)in adult yak testis in breeding and non-breeding seasons.【Method】This study used immunohistochemical SP technique to identify the distribution of VEGF and its receptor(VEGFR2),and IPP(Image-Pro Plus)statistics method was used to quantitative statistics.【Result】Immunohistochemistry showed that VEGF was expressed in the spermatogenic cells,sertoli cells,and leydig cells,and strongly expressed in the elongated sperma and vascular endothelial,no expressed in the muscle cells.VEGF'receptor(VEGFR2)was expressed in the spermatogenic cells and sertoli cells,strongly expressed in leydig cells.While weakly expressed in vessel weakly.The data showed that the average optical density of VEGF and VEGFR2 was higher in breeding season than non-breeding season in adult yak testis,but there was no significantly difference(P〉0.05).【Conclusion】VEGF and its receptors were expressed in abundance in adult yak testis,it showed that they participate in the spermatogenesis and regulate the local microenvironment.While there was certain difference in expression in different breeding season,it indicated that it closely related to the oestrous period in different seasons.
