三种核酸提取方法检测丙肝HCV-RNA定量的比较 被引量:3

摘要 目的比较及评价3种不同核酸提取方法检测HCV-RNA结果的差异。方法依照美国临床实验室标准化协会(CLSI)EP9-A2文件规定,收集40个新鲜血清标本,分别使用磁珠法、柱提法、手工法丙肝试剂以及Roche试剂进行平行检测,记录测定结果,计算线性方程及相关系数,并对其进行偏倚评估。结果磁珠法和柱提法试剂与Roche试剂方法间的相关性良好(r>0.975),而手工法与Roche试剂方法检测结果间差异超出预设允许误差范围。结论各方法学的试剂与Roche试剂检测结果大致相关性尚可,其中磁珠法结果较好。 Objective To compare and evaluate the efficacy of the three different nucleic acid extraction methods for HCV-RNA determination. Methods According to US National Clinical Laboratory Standards Committee( CLSI) EP9-A2,40 fresh serum samples were collected for parallel detection of HCV RNA reagents by magnetic beads,column extraction,manual method and Roche reagents.The results were recorded to calculate the linear equations and correlation coefficients,and estimate the bias. Results The correlation of magnetic beads method and column extraction reagent to Roche reagent were good( r 〉0. 975). Compared with Roche reagent,the expected bias of the results detected by manual method was less than the allowable error region. Conclusion Compared with other methods,the detection results of magnetic beads method are better consistent with Roche reagents.
