摘要
目的探讨高压氧能否诱导自噬产生抗神经病理性疼痛作用,从一个全新的角度验证高压氧镇痛的有效性。方法选取60只雄性SD大鼠,随机均分为4组:对照组(C组)、假手术组(S组)、坐骨神经慢性缩窄组(CCI组)和高压氧组(HBO组)。在大鼠术后7 d取脊髓标本,采用Western blot方法检测微管相关轻链蛋白1轻链3Ⅱ(LC3Ⅱ)及泛素结合蛋白P62表达,在电镜下观察各组大鼠脊髓线粒体自噬的变化情况。结果 Western blot显示CCI组、HBO组大鼠脊髓中LC3Ⅱ及P62含量较C组、S组增加(P<0.05),而C组与S组之间比较无统计学差异(P>0.05)。HBO组LC3Ⅱ表达较CCI组增加(P<0.05),而CCI组P62表达比HBO组显著增加(P<0.05)。电镜下,与C组、S组比较,CCI组、HBO组脊髓小胶质细胞的自噬明显增高(P<0.05),且C组与S组差异无统计学意义(P>0.05)。结论高压氧能诱导自噬,从而产生抗神经病理性疼痛作用。
Objective To investigate whether hyperbaric oxygen (HBO) affected neuropathic pain through autophagy and verify the HBO effect. Methods A total of 60 male SD rats were selected for the study and randomly divided into 4 groups : control group ( C group), sham operation group ( S group ), chronic constriction group (CCI group) and hyperbaric oxygen group (HBO group). LC3 Ⅱ and P62 were determined by Western blot method and the autophagy was detected by electron microscopy on the 7th day. Results CCI and HBO groups were significantly higher than C and S group in Western blot (P 〈 0.05 ). There was no difference between group C and group S (P〉0.05). The expression of LC3 Ⅱ in HBO group was significantly higher than that in CCI group (P 〈 0.05 ). And the expression of P62 in CCI group was significantly higher than that in HBO group (P 〈 0.05 ). The autophagy in CCI and HBO group were significantly increased comparing to the C and S groups (P 〈 0.05 ). However, the difference be- tween group C and S was not significant(P 〉 0.05 ). Conclusion Hyperbaric oxygen therapy can induce autophagy for anti-neuropathic pain.
