摘要
目的探讨牛初乳对感染轮状病毒乳鼠的保护作用。方法检测粪便中轮状病毒的抗原以及建立轮状病毒感染乳鼠模型,分别给予牛初乳高剂量(50g/L)、中剂量(25g/L)、低剂量(10g/L)和利巴韦林(3g/L)治疗6天,每天观察乳鼠的食欲、体重、腹泻率、腹泻严重程度及活动,同时观察小肠组织病变情况。结果乳鼠感染轮状病毒12h后均出现不同程度腹泻。模型组在第3-4天腹泻最严重,腹泻评分为3.5,且体重一直处于低水平,与正常组相比明显减轻(P<0.01);牛初乳高剂量组与利巴韦林组在第3天腹泻最严重,但腹泻分数最高2.5,低于模型组,差异显著(P<0.05),两组体重增加与模型组相比有显著差异(P<0.05),与正常组无统计学差异(P>0.05)。病理结果显示,正常对照组与牛初乳高剂量组小肠黏膜无明显肿胀、充血,黏膜下少量淋巴细胞浸润,绒毛清楚;模型组的小肠黏膜变薄,黏膜肿胀、充血,大量淋巴细胞浸润,绒毛脱落。结论牛初乳高剂量组对肠道感染轮状病毒乳鼠模型有减轻腹泻、降低腹泻率,对小肠黏膜具有修复作用。
Objective The purpose of this experiment is to make clear the bovine colostrum can protect the mice infected in rotavirus.Methods ICR neonatal mice in seven days of age were inoculated with the rotavirus via feeding tube in order to create the diarrhea model.After 12 hours,the neonatal mice emerge diarrhea and high((50g/L),middle(25g/L),low(10g/L)dose and ribavirin(3g/L)were applied in different groups,once a day,for 6days.And the appetite、weight、diarrhea rate、diarrhea severity level and movement of the neonatal mice were observed every day.Occurrence of diarrhea and death rate were detected.The small intestine was dissected into slides and stained by HE to observe the lesions of the intestinal tissue mucosal cells in Pathological changes.Results After 12 hours,the neonatal mice with infected rotavirus showed different degrees of diarrhea,model group in the 3-4days of diarrhea was obvious,and score 3.5,the weight of this group has been in low level;bovine colostrum high dose group and ribavirin group in the 3days of diarrhea were the most serious,but the highest score was 2.5,which lower than the model group,and has a significant difference(P〈0.05),the weight had a significant difference(P〈0.05)compared to the model group,while compared to the normal group with no statistical difference(P〉0.05).Pathological sections of normal group and bovine colostrum high dose group showed that the intestinal mucosa no obvious swelling,congestion,submucosal less lymphocyte infiltration,clear villi;model group of small intestinal mucosa thinning,mucosal swelling and hyperemia,lymphocytes infiltration and hair loss.Conclusion Bovine colostrum in the high dose group can relieve diarrhea、decrease diarrhea rate also can repair on the intestinal mucosa than model group in the diarrhea mucking mice.
