亚低温治疗对大鼠缺血再灌注损伤脑组织中Survivin和Bcl-2基因表达的影响 被引量:1

摘要 目的观察亚低温治疗对大鼠缺血再灌注损伤脑组织细胞中Survivin和Bcl-2基因表达的影响。方法 90只wister大鼠制作大脑中动脉闭塞(MCAO)脑缺血再灌注损伤模型。随机将大鼠分为假手术组,对照组(建模后常温处理)和观察组(建模后亚低温处理)。将对照组和观察组大鼠大脑中动脉缺血处理2,4,8小时,再灌注3h,24h,48h,7d,14d后处死。亚低温组于缺血20分钟后亚低温处理5小时。用免疫组织化学法检测Survivin和Bcl-2在缺血再灌注损伤脑组织细胞中的表达情况。结果大鼠神经功能评分:假手术组(0.15±0.04)分,对照组(3.54±0.36)分,观察组(3.72±0.55)分。亚低温处理后Survivin和Bcl-2蛋白的表达增加。结论亚低温治疗后的大鼠脑缺血组织中的Survivin和Bcl-2蛋白表达增加,对大鼠的神经细胞有保护作用,改善大鼠再灌注治疗的预后。 Objective To observe the effects of mild hypothermia therapy on regulation of cerebral ischemia reperfusion injury in rats and the expression of Survivin gene in Bcl-2tissues and cells.Methods Randomly selected 90 Wister rats are used for establish middle cerebral artery occlusion(MCAO)in cerebral ischemia reperfusion injury model.The rats were divided into sham operation group,control group(normothermic ischemia group)and observation group(mild hypothermia ischemia group).In the observation group and the control group of rats with middle cerebral artery ischemia treatment 2,4,8hours,then reperfuse 3h,24 h,48h,1week,2weeks.Mild hypothermia group reperfuse for 5hours after ischemia mild hypothermia treatment for 20 minutes.Detection of Survivin and Bcl-2cells by immune group content.Results The score of neural function in rats:sham operation group is(0.15±0.04),control group is(3.54±0.36),the observation group(3.72±0.55)points.Comparison between groups with great difference.Mild hypothermia treatment increases the expression of Survivin and Bcl 2gene.Conclusion The expression of Survivin and Bcl-2protein increased in tissue of rats after mild hypothermia treatment,and the treatment has a positive effect on rat neural cell,which can improve the prognosis of rats.
