Separation Technology of Rebaudioside Using Simulated Moving Bed Chromatography

摘要 以甜叶菊甙提取液为试验原料,研究模拟移动床色谱法分离甜叶菊甙中各因素对甜叶菊甙纯度和收率的影响。通过单因素和四元二次回归正交旋转组合试验,优化出模拟移动床色谱法分离甜叶菊甙的最佳工艺参数,即:切换时间12 min、进样质量浓度40 mg/m L、进料流速7.5 m L/min、水洗1流速10 m L/min、解吸流速15.4m L/min、再生流速7.26 m L/min、水洗2流速20.3 m L/min。各区域最优分配及连接方式:吸附区5根色谱柱三并两串,水洗1区3根色谱柱串联,解吸区6根色谱柱串联,再生区3根色谱柱一串两并,逆向进料,水洗2区3根色谱柱串联。此条件下所得甜叶菊甙纯度为80.69%,收率为93.48%。 With the extraction liquid of rebaudioside as experimental materials, the influence on the purity and yield of seperation factors of rebaudioside by simulated moving bed chromatography were studied. Used single factor experiment and orthogonal combination experiments to optimize the separation technology parameters of rebaudioside. The optimal process parameters were: switching time was 12 min, the sample concentration was 40 mg/m L, the feed flow rate was 7.5m L/min, flow rate of first washing stage was 10 m L/min, desorption flow rate was 15.4 m L/min, regeneration flow rate was7.26 m L/min, flow rate of second washing stage was 20.3 m L/min. The regional optimal allocation and connection were:three parallel and two series connection in the adsorption district, three series connection in the first washing stage, six series connection in the desorption stage, one parallel and two series connection and contrary to the feed direction in the regeneration stage, three series connection in the second washing stage. Under this operating condition, the purity of rebaudioside was 80.69%, yield was 93.48%.
