摘要
目的探讨金匮肾气丸对糖尿病(DM)大鼠肾小管细胞凋亡及蛋白Bcl-2/Bax表达的影响。方法采用小剂量高脂、高糖喂养联合腹腔注射链脲佐菌素30mg/kg造成2型糖尿病(DM)大鼠模型,金匮肾气丸溶液[800mg/(kg·d)]灌喂8周。8周后实施开腹手术取左肾组织,采用免疫组化检测肾组织Bcl—2、Bax蛋白的表达。结果与正常对照组比较,糖尿病组和金匮肾气丸治疗组肾脏细胞Bax和Bcl-2表达均增强,Bcl-2/Bax明显降低(P〈0.05),金匮肾气丸治疗组较糖尿病组Bax表达减弱,Bcl-2表达增强,Bcl-2/Bax增加(P〈0.05)。结论金匮肾气丸可能通过影响凋亡相关基因Bcl-2和Bax的表达而抑制肾脏细胞凋亡,从而发挥肾脏保护作用。
Objective To investigate the effects of Jinkui Shenqi Wan on the renal cell apoptosis in the diabetic rats. Methods Diabetic rat models were induced by high-lipids and high-sucrose feeding for a month and single intraperitoneal injection of streptozotocin ( STZ ) , then were administrated with 800 mg/( kg·d ) of Jinkui Shenqi Wan for 8 weeks.At week 8, rat left kidneys were taken and the change of Bcl-2/Bax was detected by immunohistochemistry. Results The level of Bcl-2, Bax protein and kidney tubules cells apoptos!s index were higher, while Bcl-2/Bax ratio was lower in Jinkui Shenqi Wan and DM group than those in normal control group (P〈0.05) .The levels of Bc-2 protein, Bcl-2/Bax ratio and kidney tubules cells apoptosis index were higher in Jinkui Shenqi Wan group than those in DM group ( P〈0.05 ) , and Bax expression level was reduced compared to that of DM group ( P〈0.05 ) . Conclusion Jinkui Shenqi Wan may inhibit the apoptosis of renal ceils in diabetic rats by regulating the expressions of Bcl-2 and Bax.
