以唑草·苯磺隆为模型研究酮类小分子与牛血清白蛋白的相互作用 被引量:2

摘要 在模拟动物体生理条件下,应用荧光光谱法检测Stern-Volmer猝灭常数,用紫外分光光度法检测唑草酮类除草剂对牛血清白蛋白(BSA)构象的影响,据此研究唑草酮类除草剂与BSA的相互作用机制和作用类型。用上述检测方法获得唑草酮类除草剂与BSA的猝灭常数和结合点位数,发现唑草酮类除草剂对BSA有猝灭作用。静态猝灭是唑草酮类除草剂导致BSA荧光猝灭的主要原因。加入唑草酮类除草剂后导致BSA的构象发生变化。同时用同步荧光光谱法探讨了唑草酮类除草剂对BSA构象的影响,为预防和医治除草剂中毒提供了重要依据。 Under the imitated physiological condition of animal body,the fluorescence and the spectroscopy,ultraviolet(UV)spectroscopy were employed to investigated the quenching constants and the effect of carfentrazone herbicide to bovine serum albumin(BSA),and the interaction mechanism and types between carfentrazone herbicide and BSA were studied according to this basis.The quenching constants(KSV)and corresponding combining sits digits were calculated from the spectroscopic data.After analyzing the fluorescence quenching data according to Stern-Volmer equation and Lineweaver-Burk equation,we found that carfentrazone herbicide have a quite strong ability to quench the fluorescence of BSA.The experiment results indicated that the fluorescence quenching mechanism of BSA by carfentrazone herbicide was a static procedure.The effect of carfentrazone herbicide on the conformation of BSA was analyzed by contour spectra and synchronous spectra,and some useful information was offered for preventing animals from infecting carfentrazone herbicide and curing those who had caught it.
