干扰人胃癌MGC803细胞EPCR表达对内皮细胞增殖、迁移及脉管形成的影响

摘要 目的:探讨内皮细胞蛋白C受体(endothelial protein C receptor,EPCR)在肿瘤血管形成中的作用。方法采用RNA干扰方法降低人胃癌MGC803细胞中EPCR的表达,利用Western blot 技术检测干扰EPCR效果。EPCR小干扰RNA( siRNA)片段、无关序列片段分别转染胃癌MGC803细胞,以未转染的MGC803细胞为对照;收集培养液上清,制备相应肿瘤条件培养基。人脐静脉内皮细胞( HUVECs)经不同肿瘤条件培养基作用后, CCK-8法检测HUVECs增殖能力,Transwell小室检测HUVECs迁移能力,Matrigel检测HUVECs脉管形成能力。结果与对照组及无关序列组相比,EPCR干扰组HUVECs的增殖、迁移及脉管形成能力均明显降低( P<0.05)。结论干扰胃癌MGC803细胞EPCR的表达能够抑制HUVECs增殖、迁移及脉管形成能力,提示EPCR可能在胃癌血管形成中起重要作用。 Objective To explore the effects of endothelial protein C receptor ( EPCR ) on tumor angiogenesis . Methods RNA interference was performed to silence the expression of EPCR in human gastric cancer MGC 803 cells. Then, the level of EPCR was measured by Western blotting .EPCR siRNA fragments and control fragments were transfect-ed into MGC803 cells.The supernatant of MGC803 cell media was collected to prepare tumor conditioned medium for cultivation of HUVECs .The proliferation of HUVECs was detected by CCK -8 assay.The migration of HUVECs was ex-amined by Transwell chambers .Matrigel was adopted to determine the angiogenesis of HUVECs .Results The HUVECs cultivated in media from MGC803 cells transfected with EPCR siRNA showed remarkably reduced proliferation , migration and angiogenesis than the other two groups (P〈0.05).Conclusion Down-regulated EPCR in MGC803 cells can in-hibit the proliferation , migration and angiogenesis of HUVECs , indicating the important role of EPCR in tumor angiogene-sis.
