心肌细胞中核因子-κB信号通路在雷米普利治疗心肌梗死大鼠中的作用 被引量:1

摘要 目的探讨心肌细胞中核因子-κB(NF-κB)信号通路在雷米普利治疗心肌梗死大鼠中的作用。方法利用Olivette方法进行试验分析,选取40只雄性Wistar大鼠作为研究对象。建立心肌梗死模型,随机将大鼠分为模型组和雷米普利治疗组以及假手术组。术后给予大鼠雷米普利药物灌胃治疗,灌药治疗30 d后处死大鼠,取大鼠心肌组织,然后检测大鼠的NF-κB P50、NF-κB P65、核因子κB抑制蛋白α(P-IκBα)蛋白的表达情况。结果模型组大鼠的NF-κB P50、NF-κB P65蛋白水平表达明显高于假手术组大鼠,模型组P-IκBα蛋白表达明显低于假手术组,差异有统计学意义(P<0.05);雷米普利组NF-κB P50、NF-κB P65、P-IκBα蛋白表达明显高于模型组,差异有统计学意义(P<0.05)。与假手术组相比,梗死模型组大鼠血浆ET及AngⅡ含量明显增高(P<0.05);与梗死模型组比较,雷米普利可使大鼠血浆ET及AngⅡ含量均明显降低(P<0.05)。结论心肌细胞中NF-κB信号通路在雷米普利治疗心肌梗死大鼠中具有重要的作用,NF-κB P50表达水平与心肌梗死进展有一定的相关性。 Objective To explore the role of myocardial nuclear factor- k B( NF- k B) signaling pathway in rats with myocardial infarction treated by ramipril. Methods A total of 40 male Wistar rats from Experimental Animal Center of the hospital were selected with Olivette analytic method as study subjects,the models of myocardial infarction were established according to known method,these rats were randomly divided into model group,ramipril treatment group and sham operation group. Rats in ramipril treatment group were drenched with ramipril,and they were killed in 30 days after treatment. The cardiac muscles of rats were taken out for study. Then their expression levels of NF- κB P50,NF- κB P65 and P- IκBα protein were examined. Results The expression levels of NF- κB P50 and NF- κB P65 protein in rats of model group were significantly higher than those of rats in sham operation group,the expression level of P- 1κBα protein in rats of model group was significantly lower than that of rats in sham operation group,and the difference was statistically significant( P〈0. 05). The expression levels of NF- κB P50,NF- κB P65 and P- 1κBα protein were significantly higher than those of rats in model group,and the difference was statistically significant( P〈0. 05).In comparison with the rats in sham operation group,plasma concentrations of ET and Ang II were significantly higher,and the difference was significant( P〈0. 05),and ramipril could significantly decrease the plasma concentrations of ET and Ang II in comparison with those of rats in myocardial infarction model group. Conclusion The nuclear factor- κB( NF- k B) signaling pathway in myocardial cells has important role in treatment of myocardial infarction with ramipril,and the expression level of NF- κB P50 has certain relevance with the progress and development of myocardial infarction.
