锌指蛋白217在胃癌组织中的表达及临床意义

Expression and clinical significance of zinc finger protein 217 in human gastric cancer

目的探讨锌指蛋白217（zinc finger protein 217,ZNF217）在胃癌组织中的表达及临床意义。方法收集2013年3月—2014年7月间于浙江省台州市立医院胃肠外科行胃癌根治术的58例胃癌及对应癌旁组织,同期收集临床疑似诊断为胃癌但经活检病理证实为良性疾病的胃黏膜组织30例。采用qRT-PCR检测ZNF217 mRNA在胃癌及癌旁组织中的表达,免疫组织化学染色检测ZNF217蛋白表达。收集2组患者的临床病理资料,分析ZNF217蛋白表达与胃癌患者临床及病理特征间的相关性及诊断价值。结果 ZNF217 mRNA在胃癌组织中表达水平（6.911±0.139）显著高于对应癌旁组织（1.372±0.205）,P〈0.001;ZNF217蛋白在胃癌组织中也呈现异常高表达状态（χ^2=9.910,P〈0.001）,并且胃癌组织中ZNF217蛋白高表达与肿瘤体积增大（χ^2=5.810,P=0.016）、淋巴结转移（χ^2=4.778,P=0.016）及高TNM分期（χ^2=5.806,P=0.028）呈显著正相关。ROC分析提示：ZNF217的表达作为诊断胃癌的价值较高,灵敏度和特异性均在0.8附近。结论 ZNF217 mRNA及蛋白在胃癌组织中表达上调,高水平ZNF217蛋白表达与胃癌患者恶性临床及病理特征有关。ZNF217在胃癌早期诊断及分子靶向治疗方面具有一定的科研及临床研究价值。

Objective To investigate the expression and clinical significance of zinc finger protein 217（ ZNF217） in human gastric cancer. Methods Total 58 cases of gastric cancer tissues and the corresponding tumor-adjacent normal tissues,and 30 cases of gastric mucosa which were suspected as gastric cancer but refused by biopsy pathology between March 2013 and July 2014 were collected. Real-time quantitative PCR（ qRT-PCR） was applied to detect ZNF217 mRNA expression in tissues（ n = 58）,the protein expression of ZNF217 was measured by immunohistochemistry（ IHC）. The relationship between the expression of ZNF217 and clinical features was analyzed by Pearson chi-square test. Results The expression level of ZNF217 mRNA was significantly higher in gastric cancer（ 6. 911 ± 0. 139） tissues than in adjacent normal tissues（1. 372 ± 0. 205）,P〈0. 001. The protein expression of ZNF217 was also higher in gastric cancer tissues（χ^2= 39. 910,P〈0. 001）,the high expression of ZNF217 protein was positively correlated with large tumor size（ χ^2=5. 810,P = 0. 016）,lymphatic metastasis（ χ^2 =5. 778,P = 0. 016） and advanced TNM stage（ χ^2= 4. 806,P = 0. 028）.ROC curves demonstrated that ZNF217 expression status had a high value for gastric cancer diagnosis. Conclusion The expression of ZNF217 is significantly higher in gastric cancer tissues than in tumor-adjacent normal tissues,and the upregulation of ZNF217 is associated with clinicopathological features of tumor malignance. ZNF217 may become a new marker and effective therapeutic target in early diagnosis of gastric cancer.
