摘要
目的:观察抗癌液对人大肠癌裸小鼠移植瘤p53、血管内皮生长因子(vascular endothelial growth factor,VEGF)和表皮生长因子受体(epidermal growth factor receptor,EGFR)表达的影响。方法:建立人大肠癌裸小鼠移植瘤模型,随机分为空白组、氟尿嘧啶组、抗癌液组和抗癌液与氟尿嘧啶联合用药组(简称联合组)。造模成功后第3天开始给药,连续用药7 d。采用病理学免疫组化、图像分析技术比较各组移植瘤的p53、VEGF和EGFR表达情况。结果:抗癌液能抑制移植瘤生长,无论瘤质量还是体积均明显小于空白组(P<0.01),其抑瘤作用与氟尿嘧啶大致相当,联合组抑瘤作用与氟尿嘧啶组比较亦有显著性差异(P<0.01)。抗癌液组的VEGF表达较空白组减少(P<0.05),此种作用与氟尿嘧啶作用相当,联合用药后疗效优于氟尿嘧啶,且有显著性差异(P<0.05)。抗癌液组的EGFR表达较空白组减少(P<0.05),此种作用比氟尿嘧啶稍差,联合用药后疗效显著优于氟尿嘧啶(P<0.05)。抗癌液组的p53表达较空白组增加(P<0.05),此种作用比氟尿嘧啶作用稍差,但联合用药后疗效显著优于氟尿嘧啶(P<0.05)。结论:抗癌液对人大肠癌裸小鼠移植瘤有确切的抑瘤作用,其与氟尿嘧啶的抑瘤作用大致相当。抗癌液可有效调节VEGF、EGFR和p53的表达,与氟尿嘧啶联合应用具有协同增效作用。
Objective: To observe the effect of anti-cancer liquid on human colorectal cancer nude mice transplantation tumor p53,VEGF and EGFR expression. Methods: Establishment of human colorectal cancer nude mice transplantation tumor model and the rats were randomly divided into blank control group,5-FU group,anticancer solution group and anticancer liquid and fluorouracil plus medication group. After modeling success started 3 days to medicine,7 days of continuous use. Used the pathology immunohistochemistry,image analysis technology to compare transplantation tumor in each group of p53,vascular endothelial growth factor( VEGF) and epidermal growth factor receptor( EGFR) expression. Results: The anti-cancer liquid could inhibit the growth of transplanted tumor,both from the tumor weight and volume were significantly smaller than that of blank group( P〈0. 01),the tumor suppression effect and 5- Fu roughly,joint group of tumor suppression function compared with Fluorouracil injection group also had significant difference( P〈0. 01). The anti-cancer liquid group VEGF expression of reduction in the blank group( P〈0. 05),the effect was equal to the Fluorouracil injection,curative effect is better than that of Fluorouracil injection after combination,and significant difference( P〈0. 05). The anti-cancer liquid group EGFR expression of reduction in the blank group( P〈0. 05),and this effect than Fluorouracil injection was a bit poor,curative effect was better than that of Fluorouracil injection after combination( P〈0. 05). The anti-cancer liquid group of p53 expression increased in the blank group( P〈0. 05),the less than Fluorouracil injection,but after combination curative effect was better than that of Fluorouracil injection( P〈0. 05). Conclusion:The anti-cancer liquid on human colorectal cancer nude mice transplantation tumor has exactly tumor suppression effect,its inhibitory tumor with 5-Fu roughly equivalent. The anti-cancer liquid adjust the expression of VEGF,EGFR and P53. The anti-cancer liquid associated with Fluorouracil injection applications have synergy effect.
