内异止痛汤对SD大鼠子宫内膜异位症模型在位及异位内膜NF-κB p65、VEGF干预机制的研究 被引量:13

摘要 目的:探讨内异止痛汤对SD大鼠子宫内膜异位症(endometriosis,EMs)模型在位及异位内膜中NF-κB p65、血管内皮生长因子(vascular endothelial growth factor,VEGF)的干预机制研究。方法:将40只自体移植造模成功的SD大鼠随机分为模型组、内异止痛汤低剂量组、内异止痛汤高剂量组、达那唑组以及假手术组,每组各8只,依次给予灭菌注射用水2m L·L^(-1),中药复方245.8 mg·kg^(-1),中药复方983.5 mg·kg^(-1),达那唑7.2 mg·d^(-1)以及灭菌注射用水2 m L·L^(-1)。每天灌胃1次,每次给予2 m L,连续灌胃4周,观察异位囊肿组织大小,同时采用免疫组织化学技术检测SD大鼠在位、异位以及正常子宫内膜中NF-κB p65、VEGF的蛋白表达情况。结果:1 NF-κB p65、VEGF在EMs异位内膜组织中高表达,在非EMs正常内膜组织中呈低表达,而EMs在位内膜表达居中。2经内异止痛汤干预后,与模型组比较,内异止痛汤低剂量组、高剂量组和达那唑组异位病灶体积明显减小(P<0.05);内异止痛汤高剂量组与达那唑组比较,差异无统计学意义(P>0.05)。3 QT-PCT和免疫组织化学结果:与模型组比较,内异止痛汤能下调异位内膜组织中NF-κB p65和VEGF蛋白表达P<0.05)。结论:1内异止痛汤能显著抑制大鼠腹膜异位组织生长,改善组织病理学形态变化;2内异止痛汤可以降低EMs大鼠异位组织中NF-κB p65和VEGF蛋白的表达。 Objective: To investigate Neiyi Zhitong Tang on SD rat endometriosis( EMS) model in eutopic and ectopic endometrium of NF-κB p65 and vascular endothelial growth factor vascular endothelial growth factor( VEGF) intervention mechanism research.Methods: Forty autologous transplantation made successful model of SD rats were randomly divided into 5 groups,respectively,model group,Neiyi Zhitong Tang low dose group,Neiyi Zhitong Tang high dose group and danazol group and sham operation group,each 8 rats in each,in order to give 2 mL of sterile water for injection,compound traditional Chinese medicine 245. 8 mg·kg^-1,traditional Chinese medicine compound 983. 5 mg·kg^-1,the danol 7. 2 mg·d^-1 and sterile water 2 ml·L^-1. 2 mL was administered at 4 weeks,and the expression of NF-κB p65 and VEGF in ectopic and normal endometrium were detected by using immunohistochemistry technique. Results: ① The expression of NF-κB p65 and VEGF in ectopic endometrium of Ems was lower than that in normal endometrium,and the expression of Ems in Ems. ② After Neiyi Zhitong Tang intervention,compared with the model group,ectopic lesion volume in the Neiyi Zhitong Tang low dose group,high dose group and danazol group was significantly reduced( P〈0. 05); The difference between Neiyi Zhitong Tang high dose group and danazol group was not statistically significant( P〈0. 05). ③QT-PCT and immunohistochemistry results: compared with the model group,Neiyi Zhitong Tang can cut in ectopic endometrium of NF-κB p65 and VEGF protein expression( P〈0. 05). Conclusion: ①Neiyi Zhitong Tang can obviously inhibit the growth of rat peritoneal endometriotic tissue,improve tissue pathology morphological changes; ②Neiyi Zhitong Tang can reduce the ectopic tissue of rats with endometriosis of NF-κB p65 and VEGF protein expression.
