摘要
目的观察石榴籽油对雌激素相关的骨质疏松症小鼠股骨近端骨密度及骨小梁微结构的影响,观察相关细胞的形态学变化,分析探讨其作用机制。方法将30只雌性KM小鼠按体重随机分为3组:空白对照组、实验对照组、实验组,除空白对照组实施假手术外,其余2组全部在麻醉下切除卵巢制作成骨质疏松症模型,分别给实验对照组和实验组喂食普通饲料和添加一定剂量的石榴籽油的饲料,4周后拟对小鼠股骨近端骨小梁微结构进行检测。同时对4周后3组动物采血获得其血清,利用体外血清培养成骨细胞和破骨细胞,研究3组动物血清对成骨细胞和破骨细胞相关基因表达的影响。结果实验对照组动物股骨近端骨小梁微结构破坏程度最为严重,其次为实验组,空白组骨小梁微结构正常;石榴籽油代谢物(CLA9C)抑制RANKL诱导破骨细胞的分化,通过增加碱性磷酸酶(ALP)活性和基质矿化促进成骨,同时,主要的转录水平(ALP,Coll1a1,BSP,OCN,OPG,OPN and成骨细胞标记物(DDR2,Coll1a1,ALP等)呈现上调。结论石榴子油可以抑制雌激素相关的骨质疏松症骨量的丢失同时可以促进成骨细胞分化,抑制破骨细胞活性。
Objective To study the molecular mechanism for pomegranate seed oil in prevention of bone loss in a osteoporosis mice model. Methods Accordded to the weight randomly divided 30 KM female mice into 5 groups: pomegranate seed oil low,medium and high dose group,normal group,model group,after 4 weeks detected the bone density,mineral structure and affect on uterus. Results Bone mineral density test show that low doses of pomegranate seed oil obvious decreased estrogen caused by a lack of BMD,middle dose of pomegranate seed oil completely inhibit the decline,high does of pomegranate seed oil compared with normal group,significantly increased bone mineral density,low doses of pomegranate seed oil significantly suppressed spongy bone bone loss,bone trabecular width decreased and the increase of bone trabecular spacing and was completely inhibited by middle dose of pomegranate seed oil,and high doses of pomegranate seed oil significantly increased bone mass and bone trabecular width. Mild,high doses of pomegranate seed oil inhibits a drop in the quality of the uterus. Conclusion Pomegranate seed oil can inhibit bone loss of estrogen related osteoporosis and can promote osteoblast differentiation at the same time,inhibit the osteoclast activity.
