来氟米特对大鼠肾小球系膜细胞增殖的影响 被引量:1

摘要 目的:探讨来氟米特(A771726)对大鼠肾小球系膜细胞(RMC)增殖的影响。方法:体外培养RMC,随机分为正常对照组、A771726(25μg/ml)组、A771726(50μg/ml)组、A771726(75μg/ml)组,干预培养48h后,采用CCK-8法检测各组RMC的增殖情况,流式细胞术检测各组RMC细胞周期的情况。结果:与正常对照组相比,A771726(25μg/ml)组、A771726(50μg/ml)组、A771726(75μg/ml)组RMC的增殖均被显著抑制,G1期细胞均显著增加,S期细胞均显著减少,细胞周期被阻滞于G1期(均P<0.05),并呈明显浓度依赖性。结论:来氟米特(A771726)可显著抑制RMC细胞的增殖,细胞周期被阻滞于G1期,且随着浓度的升高,其抑制作用越显著,呈明显浓度依赖性。 Objective:To investigate the effects of leflunomide(A771726)on the proliferation of the rat mesangial cells.Methods:The cultured rat glomerular mesangial cells were divided into the normal control group,A771726(25μg/ml)group,A771726(50μg/ml)group,and the A771726(75μg/ml)group.Afterr 48 hours treatment,cell proliferation was measured by cell counting kit-8(CCK-8)assay,cell cycle was measured by flow cytometry.Results:Compared with the normal control,A771726 could significantly inhibit the proliferation of rat glomerular mesangial cells,the percentage of cells with G1 phase significantly increased and the percentage of cells with S phase significantly decreased(P0.01),and its showed dose-dependently.Conclusion:Leflunomide(A771726)could significantly inhibit the proliferation and promote the apoptosis of rat glomerular mesangial cells,and its showed dose-dependently.
