摘要
利用ISSR技术对色季拉山的8个杜鹃花野生种资源进行了亲缘关系的研究。从49个引物(引物选自加拿大哥伦比亚大学UBC公司公布的100条引物)中筛选了8个条带清晰、多态性高、重复性好的引物,对8个杜鹃花野生种DNA进行扩增,共扩增出343条带(平均每个引物扩增42.88条DNA片段),多态性条带335条。用POPGENE分析数据得到8个种的遗传距离以及遗传相似性系数;利用NTSYSpc 2.1聚类分析,在遗传相似性系数约为0.55处将8种杜鹃花资源明显分为两大类,其中雪层杜鹃和柳条杜鹃被聚为第一类,其余6种为第二类。
Genetic relationship of eight species of Rhododendron located in Sejila mountain was assessed using inter-simple sequence repeat marker.Eight primers were screened out from 49 random primers,a total of 343 DNA fragments were amplified and 335 were polymorphism bands.As analyzed by POPGENE,the genetic distance among populations was with 0.377 6-0.709 4and genetic similarity coefficient was 0.491 9-0.685 5;UPGMA cluster analysis based on genetic similarity coefficient,the eight population were divided into two groups in the genetic similarity coefficient of0.55,one was the R.virgatum and R.nivale,and the other were in another group.
