摘要 目的:研究miR-20a对人炎症牙周膜干细胞成骨分化能力的影响。方法:分离培养人牙周膜干细胞后用qRT-PCR检测miR-20a的表达,并且在瞬时转染miR-20a mimics和inhibitor后成骨诱导,用茜素红染色,Western Blot和PCR的方法检测成骨能力的改变。结果:人炎症牙周膜干细胞中miR-20a表达降低,转染mimics后成骨能力升高,转染inhibitor后成骨能力降低。结论:miR-20a可以促进人炎症牙周膜干细胞成骨分化。 Objective: To study the effects of miR-20 a on the osteogenic differentiation potential of inflammatory periodontal ligament cells( IPDLSCs). Methods: Cells were isolated and cultured from the healthy and inflammatory periodontal ligament samples( HPDLSCs and IPDLSCs) respectively. miR-20 a expression was analyzed by qRT-PCR. Alizarin red staining,Western blot and PCR were used to evaluate the osteogenic differentiation potential of IPDLSCs after transient transinfection of miR-20 a mimics or inhibitor. Results: miR-20 a expression in IPDLSCs was lower than that in HPDLSCs,and the osteogenic differentiation potential of IPDLSCs were promoted by miR-20 a mimics,and reduced by miR-20 a inhibitor. Conclusion: The miR-20 a in IPDLSCs was down regulated. miR-20 a can promote the osteogenic differentiation potential of IPDLSCs.
