摘要
目的:通过Box-Behnken效应面法优化苍术-玄参药对的提取工艺。方法:以提取时间、溶媒比、浸泡时间为自变量,以总评"归一值"(包括安格洛苷C、哈巴俄苷、哈巴苷、苍术素的含量及浸膏率)为因变量,对自变量各水平进行二项式拟合,采用Box-Behnken效应面法对提取工艺优选,最后预测分析。结果:确定的最优提取工艺为:浸泡时间50 min,溶媒比11倍,提取2次,每次提取85 min。二项式拟合复相关系数较高(r=0.964 9),回归模型的预测值与实际测定值的偏差率为-2.81%。结论:Box-Behnken效应面法优选的苍术-玄参药对提取工艺,操作简便,合理可行。
OBJECTIVE To optimize extraction process of herbal pair Atractylodes Lancea-Radix Scrophulariae by Box-Behnken design.METHODS Extraction time,solvent ratio and soaking time were used as independent variables,overall desirability as dependent variable(including contents of harpagide,angoroside C,harpagoside,atractylodin and dry extract rate).Each levels of independent variables were fitted by binomial formula.Extraction process was optimized by Box-Behnken response surface method and prediction was carried out by comparing observed and forecasted results.RESULTS Optimal extraction conditions were as follows:soaking for 50 minutes by11 times of solvent,extracting for two times and85 minutes for each time.Regression coefficient of binomial fitting complex model was as high as 0.964 9.Bias between observed and predicted values was-2.81%.CONCLUSION Optimum extraction process of herbal pair Atractylodes Lancea-Radix Scrophulariae is stable,feasible and reasonable.
