不同厂家清火片中蒽醌含量的比较及聚类分析

Comparing Anthraquinone Contents in Qinghuo Tablets from Different Manufacturers and Cluster Analysis

目的:比较不同厂家清火片中大黄酚、大黄酸、大黄素、芦荟大黄素和大黄素甲醚的游离蒽醌和结合蒽醌含量,并进行聚类分析。方法:采用HPLC对10个不同厂家清火片的大黄游离蒽醌和总蒽醌含量进行测定,并运用差值法计算结合蒽醌的含量。基于系统聚类分析的基本原理,采用Ward最小方差平方和法对结合蒽醌和总蒽醌含量进行聚类分析。通过分类,将具有共性的厂家划归为一类。结果:市售10个不同厂家清火片中测得五种成分游离蒽醌含量分别为6.178,1.659,4.479,0.828,2.971,1.949,3.022,5.727,1.933,3.132 mg/g,结合蒽醌含量分别为2.282,0.468,0.429,0.496,2.404,1.055,0.304,1.424,2.446,0.830mg/g。聚类分析结果表明,类别的划分突出特性,反映蒽醌含量具有空间分布特征,不同厂家含量存在差异。结论:市售10个厂家清火片中不仅作为大黄质量控制的总蒽醌含量差别大,而且作为发挥泻下药效的主要成分结合蒽醌含量也相差悬殊。

Objective： To compare the contents of free and combined anthraquinone contents in aloe emodin,rhein,emodin,rhubarb phenol and physcion from different manufacturers Qinghuo Tablets and cluster analysis. Methods： To determine the contents of free and total anthraquinone from ten manufacturers in Qinghuo Tablet by HPLC and calculate the combined anthraquinone using the difference method. Based on the cluster analysis,use Ward minimum variance sum of squares method to analyze the combined and total anthraquinone with cluster analysis. By classification,the manufacturers in common were classified as a class.Results： Free anthraquinone contents in aloe emodin,rhein,emodin,rhubarb phenol and physcion from ten manufacturers Qinghuo Tablets were 6. 178,1. 659,4. 479,0. 828,2. 971,1. 949,3. 022,5. 727,1. 933,3. 132 mg / g and combined anthraquinone contents in aloe emodin,rhein,emodin,rhubarb phenol and physcion from ten manufacturers Qinghuo Tablets were 2. 282,0. 468,0. 429,0. 496,2. 404,1. 055,0. 304,1. 424,2. 446,0. 830 mg / g. Cluster analysis showed that the division of the outstanding characteristics which reflected the content of anthraquinone had a spatial distribution. Different manufacturers had different contents.Conclusion： Not only the total anthraquinone content which as a quality control of rhubarbvaries from commercially available ten manufacturers in Qinghuo Tablets has a big difference,but also the combined anthraquinone which plays a main ingredient of diarrhea pharmacodynamics content is different as well.
