摘要
目的:探讨化纤方对博莱霉素造模的肺纤维化模型大鼠肺系数及血清炎症因子白细胞介素-1β(IL-1β)及单核细胞趋化蛋白-1(MCP-1)的影响。方法:SD大鼠雌雄各半,随机分成空白对照组,模型组,阳性对照泼尼松组,化纤方组。除空白对照组外,其余3组均予博莱霉素制作肺纤维化大鼠模型,造模第2天起泼尼松组和化纤方组给予每日灌胃给药,分别于给药后7、11、23 d取材,分离肺组织并计算肺指数。ELISA法测定肺组织中炎症因子IL-1β及MCP-1的含量。结果:与空白对照组相比,模型组大鼠肺系数明显增加(P<0.05);与模型组比较,各给药组大鼠肺系数有不同程度的下降(P<0.01);化纤方与泼尼松比较,有统计学意义(P<0.05);模型组、泼尼松组和化纤方组血清中IL-1β含量较空白对照组明显升高(P<0.05),化纤方组血清中IL-1β含量较模型组明显降低(P<0.01);模型组和化纤方组血清中MCP-1含量较空白对照组明显增高(P<0.01或P<0.05),化纤方组血清MCP-1含量与模型组相比显著下降(P<0.01),但与泼尼松组比较明显增高(P<0.05)。结论:化纤方能够较好地控制模型大鼠肺系数的增长,并通过降低血清中IL-1β和MCP-1的含量抑制肺纤维化,对博莱霉素致大鼠肺损伤具有一定的保护作用。
Objective: To explore the effect of Huaxian Formula on lung index and inflammatory factors such as IL- 1β and MCP- 1 of rats with pulmonary fibrosis induced by Bleomycin. Methods: Healthy SD rats were randomly divided into four groups:control group,model group,prednisone( positive control) group,Huaxian formula group. The rat model was established by Bleomycin. On the next day,the two treatment groups were given drug gavage once a day while physiological saline was used as control. After 7,11 and 23 days,rats were randomly sacrificed,respectively,and the materials were drawn,observed,recorded and we analyzed lung index and inflammatory factor such as IL- 1β and MCP- 1. Results: Compared with the control group,the other groups’ lung index showed significant difference( P 〈 0. 05 or P 〈 0. 01). Compared with the model group,Huaxian formula groups’ lung index showed significant difference( P 〈 0. 01). Compared with the control group,the other groups’ inflammatory factors showed significant difference( P 〈 0. 05 or P 〈 0. 01). Compared with the model group,Huaxian formula groups’ inflammatory factors showed significant difference( P 〈 0. 05 or P 〈 0. 01). Conclusion: Huaxian formula could control lung index and reduce IL- 1β and MCP- 1. It is an effective formula in treating and preventing pulmonary fibrosis. It could intervene the injury of rats caused by Bleocamicina.
