摘要
目的探讨水陆二仙丹治疗男性畸形弱精死精子症的作用机制。方法选择健康的精子作为模型,精子悬液分为维生素C组、正常组、模型组和水陆二仙丹水提物大、中、小剂量组,剂量分别为每1mL含1.0、0.5和0.25g生药。应用次黄嘌呤-黄嘌呤氧化酶体系产生活性氧,在有氧环境下,精子悬液与0.25g/L维生素C及不同剂量(每1mL含1.0、0.5和0.25g生药)的水陆二仙丹水提物共同孵育后,采用精子低渗膨胀试验,对各组精子膜功能进行评估,同时检测各组丙二醛(MDA)水平及超氧化物歧化酶(SOD)活力。结果与模型组相比,维生素C组及水陆二仙丹水提物大、中、小剂量组1、3及6h的精子膨胀率及精子活率明显升高,差异有统计学意义(P<0.05),且水陆二仙丹水提物中剂量组(0.5g/mL)的精子膨胀率及精子活率显著高于维生素C组(P<0.05)。与模型组相比,维生素C组及水陆二仙丹水提物大、中、小剂量组MDA的水平均显著降低(P<0.05),SOD活力均显著升高,差异有统计学意义(P<0.05),且中剂量水陆二仙丹水提物(0.5g/mL)的作用明显优于维生素C组,差异具有统计学意义(P<0.05)。结论水陆二仙丹水提物具有保护精子膜功能和结构的作用,其作用与干预精子膜的脂质过氧化反应有关。
Objective To explore the mechanism of Shuilu erxiandan against male′s infertility and asthenoospermia.MethodsHuman sperm with normal physiological function were collected,and then sperm suspension were divided into the normal group,the model group,Vitamin C group and the groups of large,medium and small dose of Shuilu erxiandan.The reactive of species was made from hypoxanthine-xanthine oxidase.In the oxygen environment,Vitamin C(0.25g/L)and different content(1.0,0.5,0.25g/mL)of extract from Shuilu erxiandan were hatched with sperm.Then the function of sperm membrane in every group was evaluated using hypoosmoticswelling.Meanwhile the content of MDA and SOD were detected.Results Expansion of sperm and sperm motility in all the groups of vitamin C and large,medium and small dose of Shuilu erxiandan′s extract were significantly higher than that in the model group at time point of 1,3and 6h(P〈0.05).Also,medium dose of Shuilu erxiandan(0.5g/mL)displayed more effective on expansion of sperm and sperm motility compared with that in the vitamin group C(P〈0.05).All the groups of vitamin C and large,medium and small dose of Shuilu erxiandan′s extract significantly indicated descreased MDA content and improved SOD activity compared with that in the model group(P〈0.05).Also,this effect of medium dose of Shuilu erxiandan(0.5g/mL)on MDA and SOD activity were superior to that in vitamin C group,which showed significant difference(P〈0.05).Conclusion Extract from Shuilu erxiandan could significantly protect structure and function of sperm membrane,which is correlated with intervening its lipin peroxidation.
