4种中草药提取物对灵芝液态发酵三萜产物形成的影响 被引量:15

Effects of four kinds of Chinese herbs extracts on ganoderma triterpenoids production of Ganoderma japonicum in submerged fermentation

摘要 采用振荡-静置两段培养法,研究了以当归、党参、甘草、黄芪4种甘肃道地药材提取物作为激发子对灵芝液态发酵过程中细胞生长及产灵芝三萜的影响。结果表明:建立的最优两段发酵条件为发酵液C/N为4∶1,振荡-静置转换时间为第3天,装液量为50 m L/250 m L。在该条件下,当归、党参、甘草、黄芪提取物均能显著促进灵芝细胞生长及灵芝三萜的产生,但四者的有效作用浓度范围不同,当归、党参醇提物质量浓度在20~80 g/L促进作用良好,甘草、黄芪水提物在5~15 g/L促进作用良好。最佳添加时间分别为当归第2天,党参、甘草和黄芪均为第3天。发酵罐验证试验表明,4种中草药提取物是有效的激发子,可促进灵芝发酵三萜产量提高,尤其是当归添加量为60 g/L时,胞外三萜产量可达964.63 mg/L。 In this work,a two-stage culture process by combining shake culture with static culture was proposed to study the effects of four kinds of Chinese herbs( Angelica sinensis,Codonopsis pilosula,Glycyrrhiza uralensis and Radix astragali) on production of ganoderma triterpenoids and mycelium growth. The results showed that the optimum two-stage cultivation conditions were as follows: fermentation broth C/N was 4 ∶ 1,two-stage shift time was the 3rd day,and culture volume was 50 m L /250 m L. Under these conditions,four kinds of extracts could all significantly enhance cell growth biomass and the production of ganoderma triterpenoids,but their effective concentration ranges were different. The alcohol extracts of Angelica sinensis and Codonopsis pilosula were effective within 20 ~ 80 g/L,however,the water extracts of Glycyrrhiza uralensis and Radix astragali could enhance ganoderic triterpenoids production within 5 ~ 15 g/L. The best adding time for Angelica sinensis extract was on the 2nd day,and the others were on the3 rdday. Verification test in 1. 5 L fermentor showed that four herbs extracts were effective elicitors,which could promote the production of ganoderic triterpenoids. Especially when addition of Angelica sinensis alcohol extracts in fermentation broth was 60 g/L,the yield of extracellular triterpenoids reached 964. 63 mg/L.
