微波消融对黑色素瘤荷瘤小鼠树突状细胞抗原提呈功能的影响 被引量:1

摘要 目的研究微波消融对小鼠黑色素瘤内树突状细胞抗原提呈功能的影响。方法建立C57BL/6J小鼠B16-BL6黑色素瘤皮下移植模型,随机分成模型组、微波消融组,免疫组化检测肿瘤内CD80和CD86的表达,免疫荧光检测CD11c、MHCⅡ的表达,ELISA检测IL-12、IL-10的表达。结果微波消融组MHCⅡ、CD80和CD86表达较模型组显著增高(P<0.05);微波消融组CD11c的表达高于模型组(P<0.05);IL-12在微波消融组中高表达,而IL-10水平显著低于模型组(P>0.05)。结论微波消融可增强黑色素瘤荷瘤小鼠树突状细胞的抗原提呈功能。 Objective To explore the role of microwave ablation in antigen presentation of dendritic cells from mice bearing transplanted melanoma. Methods C57 BL / 6J mice were subcutaneously injected with B16-BL6 cells. The transplanted melanoma was treated with MA or not. The expression of CD80 and CD86 was detected by immunohistochemistry. The expressions of MHC Ⅱ and CD11 c were observed by immunofluorescence. IL-12 and IL-10 was analyzed by ELISA. Results The expression of MHCⅡ, CD80 and CD86 in mice treated with MA was significantly higher than that in model mice(P〈0.05). The mice treated with MA produced significantly more CD11 c + cells than the mice in control group. The DCs from mice treated with MA secreted more IL-12 but less IL-10 than the mice in control group. Conclusion Microwave ablation could enhance antigen presentation of dendritic cells from mice bearing transplanted melanoma.
