摘要
目的研究低水平脐血铅对新生儿端粒长度的影响及其与端粒重复序列结合因子1(TRF1)、端粒重复序列结合因子2(TRF2)的相关性。方法收集孕期无急慢性疾病的足月顺产儿脐血88例,采用原子吸收光谱法测定脐血铅浓度,以脐血铅浓度30μg/L为界分为安全浓度组(脐血铅浓度<30μg/L)48例、中毒浓度组(脐血铅浓度≥30μg/L)40例,采用全血提取基因组法提取两组脐血DNA,荧光定量PCR法测定基因组DNA的端粒长度,ELISA法测定TRF1、TRF2的浓度。结果安全浓度组端粒长度长于中毒浓度组,差异有显著性(t=2.479,P<0.05)。端粒长度与脐血铅浓度呈负相关(r=-0.301,P<0.05),TRF1与脐血铅浓度呈正相关(r=0.528,P<0.05),TRF2与脐血铅浓度呈负相关(r=-0.361,P<0.05)。结论脐血铅可通过增加TRF1表达、减少TRF2的表达来干扰新生儿端粒长度的延长,进而导致端粒长度的缩短。
Objective To investigate the effect of low level of cord blood lead on telomere length in neonates and its cor- relation with TRF1 and TRF2. Methods Cord blood was collected from 88 eases of full-term normal delivery with no history of acute or chronic diseases in pregnancy. Cord blood lead concentrations were detected using atomic absorption spectrophotometry, taking cord blood lead level of 30 μg/L as the boundary, 48 cases were classified as safe-concentration group (the cord blood lead concentrationS30 μg/L), and 40 as poisoning-concentration group (cord blood lead concentrationS30 μg/L). Employing whole blood extraction technique, cord blood DNA was extracted, the telomere length of genome DNA was measure using fluorescence quantitative PCR, and the concentrations of TRF1 and TRF2 were detected with ELISA. Results The telomere length in safe- concentration group was greater than that in poisoning-concentration group (t =2.479, P〈0.05). The telomere length was nega- tively correlated with the concentrations of cord blood lead (r=-0.301 ,P〈0.05). The TRF1 was positively correlated with cord blood lead concentration (r=0.528,P〈0.05), and TRF2 was negatively correlated (r=-0.361 ,P〈0.05). Conclusion The cord blood lead can disturb the lengthening of telomere length in neonates through increasing the expression of TRF1 and decreasing that of TRF2, resulting in shortening of telomere length.
