摘要
目的探讨骨形态发生蛋白2(BMP_2)-海藻酸钠-壳聚糖微球的制备及其促进兔桡骨骨折愈合的作用。方法采用脉冲电场法制备BMP_2-海藻酸钠-壳聚糖微球,观测其形态、尺寸,测定BMP_2载药量、包封率,并进行体外释放实验。制备新西兰兔骨缺损模型,随机分BMP_2微球组、BMP_2注射组、空微球组及空白对照组4组。于术后第30天行X线检查,比较各组骨折愈合情况。结果微球具有很好的圆形形态,平均粒径为900.79μm;微球的平均包封率为(84.97±3.49)%,BMP_2载药量平均为(16.34±0.48)%。体外模拟体液中,微球所载BMP_2被缓释出来,在第7天时87.33%的BMP_2被释放出来,在第14天时BMP_2被完全释放出来。术后第30天BMP_2微球组的骨折愈合情况较各对照组好,差异有显着性(H=6.87,P<0.05)。结论 BMP_2-海藻酸钠-壳聚糖微球具有良好的缓释效果,局部植入能促进骨折修复愈合。
Objective To study the preparation of BMP2-Chitosan-Alginate Microcasules and its effect on radius fracture healing in rabbit. Methods BMP2-Chitosan Alginate microcapsules were prepared by employing impulsive electrostatic tech- nique. Their morphology and size were observed, and encapsulation efficiency and recovery efficiency were measured. An in vitro release experiment was conducted. Models of bone defect were created in New Zealand rabbits and randomized to BMP2 microca- sules group, BMPz injection group, blank microcasules group and blank control group. An X-ray examination was done 30 days af- ter surgery. The fracture healing was compared among the four groups. Results The microcapsules had a good sphericity mor- phology with an average size of 900.79μm. The encapsulation efficiency was (84.97-3.49) %, and BMPz-loading rate of the micro capsules was (16.34± 0.48)%. BMPz was released well, and 87.33 % of BMPz was released on day 7, and completely released on day 14. On day 30 after surgery, fracture healing in BMP2 microcasules group was better than that in the other three groups, the difference was significant (H=6.87,P〈0.05). Conclusion BMP2-Chitosan-Alginate Microcapsules has a satisfactory effective- ness of delayed release. It can promote fracture healing if implanted locally.
