miRNA-16对肺腺癌A549细胞增殖和VEGF-A表达水平的影响 被引量:4

摘要 目的探讨miRNA-16作用不同时间对体外培养的人肺腺癌A549细胞增殖的影响,以及对VEGF-A表达水平的影响。方法将50 nmol/L,的miRNA-16和阴性对照序列用Lipofectamine 2000转染进人肺腺癌A549细胞中,通过qRT-PCR法检测转染后A549细胞中miRNA-16的表达水平,MTT法检测转染miRNA-16后细胞的增殖抑制情况,ELISA法检测转染miRNA-16后细胞VEGF-A表达水平。结果转染培养24h后,测得转染组miRNA-16表达水平明显高于空白对照组和阴性对照组(P<0.05);转染组细胞增殖抑制率明显高于空白对照组和阴性对照组(P<0.05);转染组细胞VEGF-A表达水平较阴性对照组和空白对照组明显降低(P<0.05)。结论 miRNA-16能引起细胞增殖抑制,并且可能与下调VEGF-A的表达有关。 Objective To observe the effects of miRNA-16 on the proliferation and expression of VEGF-A of A549 cells. Methods Human lung adenoeareinoma A549 cells were transfected with 50 nmol/L miRNA-16 mimics or negative control with Lipofectamine 2000. 24 hours after transfection, the expression level of miRNA-16 was meas- ured using qRT-PCR. Cell proliferation was measured by methylthiazol tetrazolium(MTT) assays. The expression level of VEGF-A was tested with enzyme-linked immunosorbent assay (ELISA). Results The expression level of miRNA-16 in A549 cells was significantly up-regulated compared with the blank control group and negative control group(P 〈 0. 05 ). The proliferation of A549 cells was obviously inhibited( P 〈 0. 05 ). The expression of VEGF-A in A549 cells was decreased in miRNA-16 mimics group conpared with other two groups(P 〈 O. 05 ). Conclusion The results indicate that miRNA-16 inhibits the proliferation of A549 cells, which might be associated with the down-regulation of the reduced expression of VEGF-A.
