摘要
目的探讨不同波长的色光环境对恒河猴视网膜及巩膜中视黄酸(retinoic acid,RA)含量的影响。方法 20只54 d龄的恒河猴随机分为5组(每组4只):紫光灯组、荧光灯组、白炽灯组、红光灯组及全光谱组,进行每天12 h的光照(早8点至晚8点)。于光照24周后处死,同一天上午无菌操作取材,取鼻上方视网膜及巩膜组织。取空白样品,加入全反RA标准溶液,按样品处理方法操作后,绘制标准曲线,根据标准品的色谱分析,分别确定出各组织RA的出峰位置,根据计算机的色谱工作站计算出样品的含量,以全光谱组为对照组。结果恒河猴眼视网膜、巩膜中均有RA的存在;视网膜及巩膜中RA含量间差异无统计学意义。光照24周后,红光灯组及全光谱组视网膜RA含量均高于紫光灯组、荧光灯组、白炽灯组(均为P<0.05)。红光灯组与全光谱组视网膜RA含量间差异无统计学意义;红光灯组与全光谱组巩膜RA含量高于紫光灯组,差异均有统计学意义(均为P<0.05)。余各组间两两比较差异均无统计学意义。结论恒河猴视网膜及巩膜RA含量无明显差异;视网膜中RA的含量在长波长色光环境下表达增高,在短波长色光环境下表达减少;巩膜与视网膜RA含量的变化基本一致,提示RA可能在不同色光环境下近视的形成过程中发挥一定作用。
Objective To study the differences of the retinoic acid(RA) level in the retina and sclera of rhesus monkey under different wavelengths of colored light environment. Methods Five groups of rhesus monkeys(54 days of age,4 cases in each group) were raised for 24 weeks under different wavelengths of colored light,involving purple lamp light(PL) group,fluorescent lamp light(FL) group,incandescent lamp light(IL) group,red lamp light(RL) group,and full spectrum light(FSL) group,all for 12 hours per day(from 8∶00 AM to 8∶00 PM).The nasal retinal and sclerotic tissues were collected after 24 weeks of feeding on the same day in the morning,standard solution of RA was added into blank sample,then the standard curve was drew. The peak position of RA in all tissues was found according to chromatography analysis of control standard substance. Finally,the levels of RA in samples were calculated by computer chromatography workstation. FSL group was regarded as control group. Results RA could be found in retina and sclera in the rhesus monkeys. The level of RA in sclera was similar with that of in the retina(P 〉 0. 05). After 24 weeks of feeding,the RL and FSL group had a higher RA level in retina as compared to that of PL,FL and IL groups(all P 〈 0. 05). There was no significant difference in the RA level between the RL group and FSL group(P 〉0. 05). The RL and FSL group had a higher RA level in sclera as compared to that of PL group(P 〈 0. 05). There was no significant difference between other groups. Conclusion RA can be found in retina and sclera in the rhesus monkeys. The expression of RA in the retina increase under the long wavelength light environment,and reduce when under the short wavelength light environment. The changes of RA content in the sclera are consistent with that in the retina,which indicate that RA may play the certain role in the development of myopia.
