摘要
目的研究脓毒症大鼠肺组织IL-1β和IL-4表达的变化规律,探讨右美托咪啶对脓毒症大鼠肺组织的保护作用及可能机制。方法雄性SD大鼠75只,随机分为3组:假手术组、脓毒症模型组和右美托咪啶组,每组25只,检测肺组织湿/干质量比(W/D),免疫组织化学方法和Western blot方法检测肺组织IL-1β和IL-4蛋白的表达。结果与假手术相比,急性肺损伤组的W/D比值显著升高,应用右美托咪啶治疗后,W/D比值显著降低。与对照组相比,急性肺损伤组大鼠肺组织IL-1β和IL-4蛋白表达的平均光密度值显著升高;与急性肺损伤组相比,右美托咪啶治疗组大鼠肺组织IL-1β和IL-4蛋白表达的平均光密度值显著降低。
Objective To study theexpression of IL-1β and IL-4 in the lung tissue of rats with sepsisassociated acute lung injury(ALI)to explore the dexmedetomidine protective effect on lung of rats with sepsis-associated ALI.Methods 75 male Sprague-Dawley(SD)rats were divided into:sham operation group,sepsis model group and dexmedetomidine group,25 rats in each group.The lung wet/dry(W/D)weight ratio was detected,and the expressions of IL-1β and IL-4 in lung tissue were detected by immunohistochemistry and Western blot.Results The W/D ratio of rats was higher in ALI group than in sham operation group,but decreased in dexmedetomidin group significantly than in ALI.The mean optical density values of IL-1β and IL-4 positive expressions in lung tissue were higher in sepsis model group than in sham operation group,but decreased significantly in dexmedetomidin group than in sepsis model group.Conclusion The decreased lung inflammatory reaction in septic rats by dexmedetomidine might be related to the downregulation of IL-1β and IL-4 expression.
