米糠蛋白质中氨基酸的纸上层析定性检定

摘要 采用紙上层析法鉴定米糠蛋白貿的氨基酸組成,利用茚三酮和吲醌作显色剂。吲醌与个別氨基酸作用显出了极好的,明确的特異彩色斑点,这种特異彩色借以帮助辨認RF值相近的各种氨基酸。鉴定的結果,初步确定米糠蛋白貿中含有如下的13种氨基酸:胱氨酸,精氨酸,組氨酸,天門冬氨酸,絲氨酸,谷氨酸,甘氨酸,苏氨酸,缬氨酸,苯丙氨酸,亮氨酸,丙氨酸和脯氨酸。 The amino acids in the denatured protein isolated from rice bran were inentified by paper chromatography, using ninh ydrin and isatin as developers. Isatin was found to give excellent and welldefined colour spots with individual amino acids and was used to distinguish amino acids of very close Rf values readily in our two-demersional chromatograms. Thirteen amino acids: cystine, orginine, histidine, aspartic acid, serine, glutamic acid, glycine, threonine, valine, phenylalanine, leucine, alanine and proeine were identified.
