幽门螺杆菌感染促进胃癌SGC-7901细胞CD44和CD54的表达 被引量:5

摘要 目的探讨Hp对胃腺癌SGC-7901细胞生长及肿瘤干细胞标志物CD44和CD54抗原表达的影响。方法体外建立Hp国际标准株NCTC11637感染胃腺癌上皮细胞株SGC-7901的模型,以不加Hp培养的细胞作为对照,采用光学和电子显微镜观察细胞的形态,应用流式细胞技术测定细胞CD44和CD54抗原的表达。结果在光学显微镜下,对照组细胞在培养过程中生长良好,形态规则,Hp感染组细胞在感染10h左右出现细胞脱壁现象,随着感染时间的延长,细胞发生空泡样病变、胞浆破裂等形态学改变;电镜观察显示,对照组细胞细胞膜表面微绒毛、细胞器和细胞核形态均正常,Hp感染组细胞出现微绒毛消失、细胞浆空泡样病变、细胞核裂解等细胞损伤现象。Hp感染组细胞的CD44抗原表达荧光强度为(6.45±0.38)fmol/m L,高于对照组(4.27±1.27)fmol/m L,差异有统计学意义(P<0.05);Hp感染细胞的CD54抗原表达荧光强度为(34.00±4.50)fmol/m L,高于对照组(3.13±0.06)fmol/m L,差异有统计学意义(P<0.01)。结论 Hp感染可直接导致胃上皮细胞形态发生损伤,同时促进感染胃黏膜细胞肿瘤干细胞标记物CD44和CD54抗原的表达。 Objective To explore the effect of Helicobacter pylori(H. pylori) on the growth of gastric epithelial cells andthe expression of CD44 and CD54 in gastric epithelial cells, SGC-7901, in vitro. Methods Cultured SGC-7901 cells wereinfected by H. pylori NCTC11637 with uninfected cells as a normal control. Morphological changes of cells were observed usinglight microscope(LM) and transmission electron microscope(TEM). Expression of CD44 and CD54 surface antigens wereexamined by flow cytometry(FCM). Results Under LM, control gastric epithelial cells grew well in the culture with a regularmorphology. Whereas 10 hours post infection, the H. pylori-infected cells started to detach from the wall, and demonstratedvacuolar changes, cell rupture and endochylema leakage.Under TEM, the control cells showed normal morphology withmicrovilli on the membrane, organelles and cell nuclei while the infected cells showed disappearance of microvilli, cytoplasmicvacuolar changes and nucleus split. Compared with the control group, the expression of both CD44 and CD54 antigens on theSGC-7901 cells in the infection group was enhanced(t=4.028, P〈0.005; t=16.802, P〈0.001). Conclusion Infection of H.pylori could directly cause morphological changes of gastric epithelial cells, and promote the expression of CD44 and CD54 proteins, which may facilitate the development of gastric carcinoma.
