槲皮素对敌敌畏诱导肺毒性影响研究 被引量:1

Effects of Quercetin Against Dichlorvos Induced Lung Toxicity on Rats

摘要 目的研究槲皮素对敌敌畏诱导的大鼠肺毒性的影响。方法将80只雄性Wistar大鼠按体重随机分为8组:正常对照组,槲皮素低剂量组,槲皮素中剂量组,槲皮素高剂量组,敌敌畏处理组,低剂量槲皮素与敌敌畏联合组,中剂量槲皮素与敌敌畏联合组,高剂量槲皮素与敌敌畏联合组。槲皮素经灌胃给药,敌敌畏通过饮水给予大鼠,连续给药90 d。检测肺脏组织中超氧化物歧化酶(SOD),过氧化氢酶(CAT),谷胱甘肽过氧化物酶(GPx)的活性及丙二醛含量(MDA),并观察肺脏组织病理学变化。结果与对照组大鼠相比,敌敌畏组大鼠肺脏组织丙二醛(MDA)含量及抗氧化物酶类活性显著升高(P<0.01)。当大鼠同时给予敌敌畏和槲皮素后,上述指标均有下降趋势。与敌敌畏组相比,敌敌畏与高剂量槲皮素组大鼠肺脏组织MDA含量显著下降,且超氧化物歧化酶(SOD)、过氧化物酶(CAT)和谷胱甘肽过氧化物酶(GPx)活性显著降低(P<0.05)。上述指标在3个剂量的单独槲皮素组与对照组相比没有统计学差异(P>0.05)。肺脏病理组织学检测结果显示,敌敌畏组大鼠肺脏组织可见肺泡间质增厚、单核细胞浸润、肺气肿;这种病理学改变在高剂量槲皮素与敌敌畏联合组有所改善。结论槲皮素对敌敌畏所诱导的肺毒性具有部分保护作用。 Objective To study the effects of quercetin against dichlorvos induced lung toxicity on rats. Methods Eighty male Wistar rats were randomly divided into eight groups: control group,low- dose quercetin group,middle-dose quercetin group,high-dose quercetin group,dichlorvos group,low-dose quercetin plus dichlorvos group,middle-dose quercetin plus dichorvos group,high-dose quercetin plus dichlorvos group. Quercetin was given to rats by gavage and dichlorvos was given to rats by drinking water continually for 90 days. The activities of superoxide dis mutase( SOD),catalase( CAT),glutathione peroxidase( GPx) and the content of malondialdehyde( MDA) in lung tissues of rats were measured,histopathological changes in lung tissues were observed as well. Results Co mpared with the control rats,the content of MDA and activities of SOD,CAT and GPx were significantly increased in dichlorvos group( P〈0. 01). When the dichlorvos was given to rats together with quercetin,the afore mentioned indices showed downward trend. Co mpared with the dichlorvos group,the content of MDA and activities of SOD,CAT,GPx were significantly decreased in high- dose quercetin plus dichlorvos group( P〈0. 05). While co mpared with the control group,the afore mentioned indices did not show significantly changes in quercetin alone treated groups( P〉0. 05). Histological exa mination showed thickened alveolar interstitial,infiltration of mononuclear cells and e mphyse matous in dichlorvos treated lung tissues,and these changes were milder in high-dose quercetin plus dichlorvos treated rats. Conclusion Quercetin at 50 mg / kg. bw could partly prevent the lung toxicity induced by dichlorvos.
