超声波辅助双酶法提取假酸浆总黄酮及抑菌性检测 被引量:3

摘要 采用超声波-双酶法提取假酸浆中总黄酮,考察纤维素酶和果胶酶共同作用时酶解时间、酶解温度、pH值以及酶用量对假酸浆中总黄酮提取的影响,考察假酸浆黄酮对几种病菌的抑制作用。结果表明:超声波辅助双酶法提取假酸浆黄酮的最佳工艺条件是:pH值4.6、酶解温度55℃、加酶比(果胶酶:纤维素酶)3∶1、酶解时间70min,此条件下假酸浆中总黄酮的提取率是5.63%。假酸浆中总黄酮对大肠杆菌、青霉菌、沙门氏菌、和枯草芽孢杆菌有明显的抑制作用,最低抑菌浓度分别为:1.25、2.5、2.5、5mg/mL。 To extract total flavonoids by ultrasonic-double-enzumatic method,using nicandra physaloides as raw material,we investigated the effect of enzymolysis time,enzymolysis temperature,pH value and enzyme dosage on total flavonoids of nicandra physaloides extraction,together with combined effect of cellulase and pectinase.Also,the inhibition of some germs by flavonoids of nicandra physaloides was investigated in details.The results show that,the optimum technological conditions for flavonoids of nicandra physaloides extraction by ultrasonic-double-enzumatic method are as follows:enzymolysis temperature 55℃,pH value4.6,enzymolysis time70 min,enzyme ratio(pectinase:cellulase)3∶1,and the extraction ratio of the total flavonoids of nicandra physaloides under the conditions is 5.63%.The total flavonoids of nicandra physaloides have obvious inhibition on escherichia coli,penicillium,salmonella and bacillus subtilis In additon,the minimal inhibitory concentrations are 1.25,2.5,2.5,5mg/mL respectively.
