摘要
目的观察健脾补血方对Lewis肺癌小鼠化疗后血清中5羟色胺(5-hydroxytryptamine,5-HT)、P物质(Substance P,SP)以及5羟色胺3A受体(5-hydroxytryptamine 3A receptor,5-HT3AR)、速激肽1受体(Neurokinin1 receptor,NK1R)的影响,探讨健脾补血方对化疗后恶心呕吐的改善作用。方法 C57BL/6小鼠腋下接种Lewis肺癌细胞,随机分为肺癌组,模型组,健脾补血方高、中、低剂量组。模型组及健脾补血方各剂量组连续尾静脉注射顺铂3 d(4,3,3 mg·kg^(-1))复制模型。健脾补血方高、中、低剂量组分别给予健脾补血方25,12.5,6.25 g·kg^(-1)生药量,肺癌组和模型组给予生理盐水,连续灌胃给药14 d,末次给药后禁食24 h取血液标本及脑组织,酶联免疫吸附法(ELISA)检测血清中5-HT和SP的浓度,并采用蛋白印迹法(Western Blot)检测5-HT3AR和NK1R的蛋白表达。结果与肺癌组比较,模型组血清中5-HT和SP浓度明显升高,5-HT3AR和NK1R的蛋白表达量升高;与模型组比较,健脾补血方各剂量组可降低血清中5-HT及NK1R蛋白表达量,而健脾补血方高剂量组可降低SP浓度及5-HT3AR蛋白表达量。结论健脾补血方能降低Lewis肺癌小鼠化疗后血清中5-HT和SP浓度,并下调其受体5-HT3AR和NK1R的蛋白表达量,可能与抑制化疗后恶心呕吐因子表达有关,从而减轻化疗后恶心呕吐反应。
Objective To observe the effects of Jianpi Buxue Decoction(JBD) on 5- hydroxytryptamine(5- HT),substance P(SP) and their receptors 5- hydroxytryptamine 3A receptor(5- HT3AR) and neurokinin1 receptor(NK1R)in Lewis lung cancer mice treated with chemotherapy, and to investigate the effects of JBD on improving nausea and vomiting after chemotherapy. Methods After adaptively fed for seven days, the C57BL/6 mice were inoculated with Lewis lung cancer cells into the armpit,and then were randomly divided into lung cancer group, model group, and high-, medium and low- dose JBD groups. Model group and JBD groups were injected with cisplatin(4,3,3 mg·kg^(-1))through tail vein to induce Lewis lung cancer. The mice of lung cancer group and model group were given saline orally,and JBD groups were given 25,12.5,6.25 g·kg^(-1) of JBD orally for 14 continuous days respectively. After the last medication,all the mice were starved for 24 hours and then were executed. The blood samples and brains were collected. ELISA method was used to detect the concentrations of 5- HT and SP in serum, and Western blot method was used to detect the concentrations of 5- HT3 AR and NK1 R. Results Compared with the lung cancer group, the concentrations of 5- HT, SP in serum and the protein expression of 5- HT3 AR and NK1 R in the model group were increased significantly. Compared with the model group,the concentrations of 5- HT in serum and NKIR expression in JBD groups and the protein expression of 5- HT3 AR and serum SP in high- dose JBD group were decreased significantly. Conclusion JBD could significantly decrease the concentrations of 5- HT, SP in serum and the protein expression of 5- HT3 AR and NK1 R in Lewis lung cancer mice after chemotherapy,which contribute to inhibiting the factor expression of nausea and vomiting after chemotherapy,thus to relieve nausea and vomiting.
