女贞苷对Aβ42诱导的神经细胞凋亡及相关蛋白NF-κB、Bcl-2变化的影响 被引量:4

Effect of Nuezhenoside on Aβ42-induced SH-SY5Y Cell Apoptosis and Apoptosis-related Protein NF-κB and Bcl-2 Expression

摘要 目的探讨女贞苷对Aβ42诱导的神经细胞凋亡及相关蛋白NF-κB、Bcl-2变化的影响及其相关机制。方法人神经母细胞瘤细胞(SH-SY5Y细胞)用女贞苷(50,100μmol·L^(-1))预保护12 h后,加入浓度为15μmol·L^(-1)的Aβ42诱导损伤12 h,用MTT法测定终点细胞存活率,Western blot法检测NF-κB和Bcl-2的蛋白表达变化。结果 50,100μmol·L-1的女贞苷可减少Aβ42引起的SH-SY5Y细胞凋亡。女贞苷组NF-κB的蛋白表达较模型组减少(P<0.01),抗凋亡因子Bcl-2的蛋白表达相对于模型组明显增加(P<0.01)。结论表明女贞苷可提高Aβ42诱导损伤的SH-SY5Y细胞存活率,其机制可能与女贞苷抑制NF-κB的激活和增加抗凋亡因子Bcl-2的蛋白表达有关。 Objective To observe the protective effect of nuezhenoside(ligustroflavone) on Aβ42- induced SH- SY5 Y cell apoptosis and on the expression of NF- κB and Bcl- 2. Methods Human neuroblastoma SH- SY5 Y cells were pretreated with nuezhenoside at various concentrations(50,100 μmol/L) in serum- free medium for 12 hours at 37 ℃and then were incubated with 15 μmol·L of Aβ42 for another 12 hours. Cell viability was measured by MTT. The expression of NF-κB and Bcl- 2 was detected by Western blot. Results Nuezhenoside(50,100 μmol·L)significantly increased the survival rate of SH- SY5 Y cells. Western blot results showed that the expression of NF-κB was lower and Bcl- 2 expression was higher in both nuezhenoside groups than the model group(P 〈0.01). Conclusion The results indicated that nuezhenoside can obviously protect SH- SY5 Y from Aβ42- induced cell apoptosis, and its possible protective mechanism is related with the inhibition of NF- κB activation and with the increase of Bcl- 2 protein expression.
