促性腺激素释放激素类似物逆转卵巢恶性生殖肿瘤对顺铂的耐药性研究被引量：1

摘要目的观察促性腺激素释放激素类似物曲普瑞林逆转卵巢恶性肿瘤对顺铂耐药性的效果。方法建立卵巢肿瘤顺铂耐药细胞系模型OVCAR-3/CDDP,采用MTT比色法分别检测单独使用曲谱瑞林、单独使用顺铂、曲谱瑞林和顺铂联用时对OVCAR-3/CDDP细胞的抑制效果,根据血药峰值浓度的不同分成顺铂组、曲谱瑞林组、两药联合组和OVCAR-3组。观察各组的细胞生长情况,分析曲谱瑞林对OVCAR-3/CDDP细胞顺铂耐药性的逆转作用。结果顺铂组、曲谱瑞林组、OVCAR-3组细胞的生长状况相近,细胞在不同时间点的生长抑制率差异无统计学意义(P>0.05);而两药联合组的细胞生长抑制率与其他3组比较,差异有统计学意义(P<0.05)。与顺铂组、曲谱瑞林组相比,顺铂和曲谱瑞林联用组能更显著地削弱OVCAR-3/CDDP细胞的耐药性。结论曲谱瑞林能在一定程度上抑制卵巢肿瘤顺铂耐药细胞的生长,逆转卵巢肿瘤顺铂耐药细胞系的耐药性。
