摘要
[目的]观测补脾益气中药对脾气虚痴呆大鼠水迷宫实验、血清白介素1、白介素6影响。[方法]使用随机平行对照方法,将32只SPF级SD大鼠按随机数字表分为4组,空白组,痴呆模型组、脾气虚痴呆组,补脾益气组,8只/组。使用饮食不节加劳倦伤脾复合因素方法复制脾气虚模型,实验第1d,脾气虚痴呆组和补脾益气组大鼠单日游泳至耐力极限,禁食饲料,喂饲甘蓝(20~30g/只);双日喂饲猪油脂每只2mL/100g;连续15d。在此基础上使用氧化铝痴呆造模方法建立氧化铝痴呆模型,脾气虚模型造模同时,痴呆模型组、脾气虚痴呆组,补睥益气组均灌服氧化铝500mg/kg,1次/d,连续90d。灌胃干预,补脾益气组:实验16d开始,补脾益气中药(人参、白术各20g,黄芪40g,当归20g,升麻6g,柴胡、橘皮各12g,甘草6g),1.5mL/100g,连续75d;其他各组等量生理盐水。检测水迷宫学习记忆能力(定位航行时间,Morris水迷宫实验)、血清IL-1、IL-6(酶联免疫法)。[结果]实验第15d(造模15d),脾气虚痴呆组和补脾益气组大鼠表现为倦怠、懒动、蜷缩聚堆、眯眼、弓背,粪便时干时软,偶有便溏,食量与饮水量均减少,脾虚模型复制成功。实验第95d水迷宫学习记忆能力定位航行时间痴呆模型组、脾气虚痴呆组、补脾益气组均明显高于空白组(P〈0.01),脾气虚痴呆组明显高于痴呆模型组(P〈0.01),补脾益气组明显低于痴呆模型组和脾气虚痴呆组(P〈0.01)。实验第96d血清IL-1、IL-6痴呆模型组、脾气虚痴呆组、补脾益气组均明显高于空白组(P〈0.01),睥气虚痴呆组明显高于痴呆模型组(P〈0.01);补脾益气组明显低于痴呆模型组和脾气虚痴呆组(P〈0.01)。[结论]补脾益气中药可提升脾气虚痴呆大鼠水迷宫实验学习记忆能力(定位航行时间)、降低血清白介素1、白介素6。
[Objective]Observation of spleen Qi of traditional Chinese medicine on senile dementia of deficiency of spleen of rats with water maze test,and serum Interleukin 1,Interleukin-6 impact.[Methods]32rats were randomly divided into 4 groups by random parallel control method.Establish the animal models of spleen deficiency by classical approach.On this basis,establish the alumina-dementia model and intervene with traditional Chinese medicine prescriptions of invigorating spleen and replenishing Qi.And compare the changes of the rats' learning and memory capabilities according to the behaviors changes by Morris water-maze.Detect the IL-1 and IL-6 concentrations in serum of each group rats by Enzyme-linked-immunosorbent-assay.[Result]Positioning time testing in rats,spleen were significantly higher than the model group(P〈0.01).Invigorating the spleen and deficiency of spleen-Qi group was significantly lower than the model group groups(.P〈0.01).Serum IL-1,IL-6,spleen were significantly higher than the model group(P〈0.01).Invigorating the spleen and deficiency of spleen-Qi group was significantly lower than the model group groups(P〈0.01)[Conclusion]The treatment of invigorating spleen and replenishing Qi could effectively improve the rats' learning and memory capabilities and decrease the IL-1 and IL-6 concentrations in serum.
