摘要 目的:探讨8-羟鸟嘌呤对小鼠骨骼肌成肌细胞C2C12增殖分化的影响。方法:基于C2C12增殖分化模型,通过有无添加8-羟鸟嘌呤,建立实验组和对照组。显微镜观察各时期细胞形态,对其各个时期进行细胞计数;检测两组细胞成肌相关分子Myo D、M-Cadherin和Myo G的m RNA表达。结果:与对照组相比,在细胞增殖阶段,实验组的分裂相和细胞数目明显多于对照组(P<0.01);而在细胞分化阶段,实验组的Myo D、M-Cadherin和Myo G的m RNA表达增强(P<0.05或P<0.01)。结论:8-羟鸟嘌呤可能对小鼠骨骼肌成肌细胞C2C12增殖和分化有促进作用。 Objective To investigate the effects of 8-oxoguanine(8-oxo G)on the proliferation and differentiation of mouse skeletal muscle C2C12 cells. Methods The experimental group and control group were established using C2C12 proliferation and differentiation model. 8-oxo G was extra added to the experimental group. The morphology and number of C2C12 cells,and the PCR expression of Myo D,M-Cadherin and Myo G were observed throughout the experiment. Results The number of C2C12 cells in proliferation stage increased significantly and the expression of Myo D,M-Cadherin and Myo G in differentiation stages enhanced significantly in experimental group as compared with the control group(P〈0.05 or P〈0.01). Conclusion 8-oxoguanine effectively promotes proliferation and differentiation of C2C12 cells.
