摘要 目的研究Wnt信号通路中β-catenin、TCF-4和Wnt-2在非小细胞肺癌中的活化状态及其与靶基因VEGF和Survivin表达的相关性,探讨Wnt信号通路对非小细胞肺癌细胞增殖、侵袭转移以及对靶基因表达的影响,为非小细胞肺癌多靶点基因治疗提供实验依据。方法应用RT-PCR、免疫组化和Western blot等方法,检测非小细胞肺癌的癌组织及癌旁正常组织中β-catenin、TCF-4和Wnt-2的表达,以及在Wnt信号通路的活化状态下靶基因VEGF和Survivin表达情况。结果在非小细胞肺癌组织中,β-catenin、TCF-4和Wnt-2的表达明显高于癌旁正常组织(P<0.01);Wnt^-组与Wnt^+组在癌旁正常组织和NSCLC组织中分布差异具有统计学意义(P=0.000);VEGF和Survivin在Wnt^+组中的表达频率均明显高于Wnt^-组。结论非小细胞肺癌组织中Wnt信号通路被活化,其通路活化可能是导致非小细胞肺癌各靶基因表达增加的重要因素,并与非小细胞肺癌的发生、发展密切相关。 Objective To study the correlation between the status of the aviation of Wnt-2 and β-catenin/TCF-4 and the expression of its target protein VEGF and Survivin, to discuss the influence of the expression of Wnt signaling pathway to the proliferation and apoptosis of NSCLC cells. Methods Immunohistochemistry, RT-PCR and western blotting were used to detect Wnt signaling activation and its correlation of its target protein expression inNSCLC tissue, adjacent tissue and normal tissue. Results Expressions of Wnt-2, β-catenin and TCF-4 were significantly higher in cancer tissues compared with normal tissues (P〈0.01). The distribution of Wnt+and Wnt-group in normal and cancerous tissues were significantly different (P=0.000). In Wnt+ group,the expression of VEGF and Survivin were significantly higher than the Wnt-group. Conclusion Wnt signaling activation maybe a major factor of the expression the target protein which plays a important role in carcinogenesis of NSCLC.
