摘要
〔目的〕测定小鼠创伤后N-cadherin蛋白的表达,探讨N-cadherin在创伤修复中的作用。〔方法〕用皮肤切除法在小鼠背部建立1 cm2小鼠皮肤创伤模型,分为复方组,腺嘌呤组,模型组和正常对照组。复方组小鼠给予中药复方制剂[附子(制)∶三七∶紫草=1∶2∶3];腺嘌呤组给予腺嘌呤;模型组不给药。实验结束后,观察创伤面积及修复的快慢程度变化。创伤组织皮肤提取蛋白,Western blot法测定皮肤组织EMT过程中N-cad在创伤修复中表达的程度高低。〔结果〕复方组小鼠背部创伤恢复最快,其次为模型组,腺嘌呤组小鼠创伤愈合最慢;Western blot法测定显示,创伤后N-钙调素的表达从高到低依次为:腺嘌呤组、模型组、复方组,与正常对照组比较其差异具有统计学意义(P<0.05)。〔结论〕复方组给予的中药复方制剂对小鼠创伤的愈合有促进作用,腺嘌呤组给予腺嘌呤造成小鼠阳虚模型,不利于小鼠创伤愈合;小鼠创伤后N-cad的表达越低,创伤修复越快。
〔Objective〕Investigating the expression of N-cadherin in wounded mice and deter minating the action of N-cadherin in the process of wound healing. 〔Methods〕1-cm-diameter skin wounds were inflicted on the back of mice, and divide into four groups: the group of Compound; the group of Adenine; the group of model; the group of blank. The group of compound was treated with compound preparation( monkshood: pseudo-ginseng: radix lithospermum = 1 ∶ 2 ∶ 3); the group of Adenine was treated with Adenine; the group of model was treated nothing. After the experiment,observing the changes of wound area and speed of wound repair. Extracting the protein of the wound skin and investigating the level of N-cadherin of the wound skin in the process of EMT by western blot. 〔Results〕The speed of wound healing in compound group is the fastest,followed by the group of model,and the group of Adenine is the slowest. It shows that the expresstion of N-cadherin statue is : the group of Adenine is the highest, the group of model is higher and the group of compound is the lowest comparing with the control group( P〈0. 05).〔Conclusion〕The compound preparation is useful to wound healing. Adenine can resist wound healing. After injured,the lower Ncadherin's expresstion the faster wound repaired.
