摘要
目的:从慢性阻塞性肺疾病(COPD)模型大鼠不同时间段红细胞数(RBC)、血红蛋白(Hb)、白细胞数(WBC)、促红细胞生成素(EPO)、转化生长因子(TGF-β1)及基质金属蛋白酶-9(MMP-9)几个关键环节,探讨COPD气道重塑与血常规、EPO的相关性,推测合并贫血的可能性。方法:采用气管内滴注脂多糖加烟熏法建立大鼠COPD模型,分别设正常组、COPD模型4周组、COPD模型8周组,造模结束后对大鼠分别进行血液和肺泡灌洗液(BALF)的采集,并对相关指标进行检测。结果:RBC、Hb、EPO在4周组中均有不同程度增高,但在8周组则有不同程度的下降;TGF-β1、MMP-9、WBC则在两个时间段中均有不同程度的升高。相关性分析:EPO与RBC、Hb成正相关;4周组和8周组TGF-β1、MMP-9与WBC成正相关;8周组RBC、Hb、EPO与TGF-β1、MMP-9成负相关。结论:随着COPD病情进展,气道重塑重要细胞因子与血液主要指标的相关性越明显,出现贫血合并症的可能性越大,可以尝试从中医药角度探索干预策略或途径。
Objective: To explore the correlation of COPD airway remodeling between routine blood and EPO from red blood cells( RBC),hemoglobin( Hb),white blood cell count( WBC) and erythropoietin( EPO),transforming growth factor( TGF-beta 1) and matrix metalloproteinases 9( MMP-9).Methods: The endotracheal drip lipopolysaccharide with smoked method was used to establish the rat COPD model,respectively setting up normal group,COPD model group,COPD model 4 weeks group,COPD model 8 weeks group.4 weeks after the building modeling,blood and alveolar lavage fluid( BALF) were collected and the related indicators were tested.Results: RBC,Hb and EPO in 4 weeks group were increased in different degrees and those in 8 weeks group decreased in different degrees.TGF-β1,MMP-9 and WBC were increased in two time periods.Correlation: EPO,RBC and Hb were positively related; TGF-β1,MMP-9 and WBC in 4 and 8 weeks groups were positively related.RBC,Hb and EPO and TGF-β1and MMP-9 in 8 weeks group were negatively related.Conclusion : With COPD progression,airway remodeling important cytokines,the more obvious correlation with blood main indicators and anemia,the higher the risk of complications.We can try to get TCM perspective to explore the intervention strategy or approach.
