胃癌组织中幽门螺旋杆菌感染与SOX2、CyclinD1蛋白表达的相关性研究 被引量:15

摘要 目的探讨胃癌组织中幽门螺旋杆菌(HP)感染与SOX2、细胞周期蛋白D1(CyclinD1)蛋白表达的相关性。方法收集2014年1月—2015年1月河北北方学院附属第一医院经胃镜和病理检查明确诊断的胃癌标本50例,根据是否感染HP分为HP阳性组(36例),HP阴性组(14例)。采用^(13)C呼气试验和吉姆萨染色(Giemsa)法检测幽门螺旋杆菌感染,采用免疫组织化学法标记SOX2、CyclinD1,检测其在胃黏膜组织中的蛋白表达情况。比较胃癌组织SOX2、CyclinD1蛋白表达与胃癌临床病理特征之间的关系及与HP感染的相关性。结果 SOX2蛋白的表达水平与胃癌患者的性别、年龄和肿瘤大小无关(P>0.05),与临床分期、浸润深度和分化程度有关(P<0.05);CyclinD1蛋白的表达水平与胃癌患者的性别、年龄和肿瘤大小无关(P>0.05),与临床分期、浸润深度和分化程度有关(P<0.05)。SOX2阳性颗粒定位于细胞核,呈棕黄色颗粒。SOX2蛋白在HP阳性组胃癌组织中阳性表达率为5.6%,在HP阴性组阳性表达率为35.7%,2组比较差异有统计学意义(x^2=5.316,P<0.01)。CyclinD1主要定位于胃癌细胞的细胞核,呈棕黄色颗粒。HP阳性组胃癌组织中CyclinDl蛋白阳性表达率为66.7%,HP阴性组表达率为28.5%,2组比较差异具有统计学意义(X^2=2.119,P<0.05)。胃癌组织中HP感染与SOX2蛋白表达呈负相关(r=-0.81l,P<0.01),与CyclinD1蛋白表达呈正相关(r=0.274,P<0.05)。结论 HP感染可能诱发胃癌,SOX2、CyclinD1蛋白可能参与胃癌的发生、发展过程。 Objective To investigate the relationship between the Helicobacter pylori(HP) infection and the expression of SRY related transcription factor 2(SOX2) whichwas similar to that of box HMG and cyclin Dl(CyclinDl) protein expression.Methods Collect from January 2014 to January 2015,in Hebei North University First Affiliated Hospital by gastroscopy and pathology diagnosis of gastric cancer specimens of 50 cases,according to whether HP infection,they were divided into HP positive group(36 cases),HP negative group(14 cases).The 13 C breath test and Giemsa staining(Giemsa) method for detection of H.pylori infection,using immunohistochemistry markers SOX2,CyclinDl to detect its protein in gastric mucosa tissue expression.Compared the relationship between the expression of SOX2 and CyclinDl protein in gastric carcinoma and the clinicopathological characteristics of gastric carcinoma and the correlation with HP infection.Results The levels of SOX2 protein expression in patients with gastric cancer has no relation with sex,age and tumor size(P〉0.05),and related with clinical stage,invasion depth and degree of differentiation(P〈0.05);CyclinDl protein expression levels of patients with gastric cancer has no relationship with sex,age and tumor size independent(P〉0.05),but related with clinical stage,tumor invasive depth and differentiation(P〈0.05).SOX2 positive particles were located in nuclei,brownish yellow granules.HP positive gastric carcinoma tissue SOX2 protein positive expression rate was 5.6%,in HP negative group,the positive expression rate was 35.7%,the difference between the two groups has statistical significance(x^2 =5.316,P〈0.01).CyclinDl mainly located in the nucleus of gastric cancer cells,brownish yellow granules.HP positive group' s gastric carcinoma CyclinDl protein expression rate was 66.7%,HP negative group' s expression rate was 28.5%.The differences between the 2 groups were statistically significant(x^2 = 2.119,P〈0.05).HP infection in gastric carcinoma was negatively correlated with S0X2 protein expression(r = — 0.811,P〈0.01),and was positively correlated with CyclinDl protein expression(r- 0.274,P〈0.05).Conclusion The HP infection may induce gastric cancer,SOX2,CyclinDl protein may be involved in the occurrence and development of gastric cancer.
