摘要
目的:探讨附子理中汤干预脾肾阳虚型溃疡性结肠炎(Ulcerative colitis,UC)可能的药效环节。方法:SPF级大鼠48只按随机数字表法分为正常对照组、模型组、柳氮磺胺吡啶组、附子理中汤高、中、低剂量组6组。中西药联合制备符合脾肾阳虚证的免疫化学性复合模型,药物灌肠干预后用ELISA法检测IL-6、IL-8在血清中的含量,取结肠组织,免疫组化染色(SP)法检测ICAM-1的表达,并观察其病理组织学情况。结果:中药各剂量组与模型组比较,血清中IL-6、IL-8水平、结肠黏膜病理评分及其ICAM-1表达均有所下降,各项检测指标表明附子理中汤高剂量组的疗效优于柳氮磺胺吡啶组,差异有统计学意义。结论:附子理中汤对脾肾阳虚型UC疗效确切,作用机制可能与降低血清促炎症因子及细胞间黏附因子的表达有关。
Objective:To explore the possible mechanism of the Fuzi Lizhong decoctions therapeutic effect on spleen- kidney- yang -deficiency type in rats with ulcerative colitis. Methods: Forty-eight rats were randomly divided into control group, model group, sulfasalazine group, high-dose, medium-dose and low-dose group of Dandelion extract. Immune chemical compound model ,which conforms to the spleen kidney yang deficiency syndrome are made by combing western and traditional Chinese medicine. After giving medicine, ELISA was performed to detect the levels of interleukin-6 (IL-6), interleukin-8 ( IL-8 ) and Immunohistochemical and SP method was performed to detect the expressive levels of ICAM-1. The histological changes of mucous membrane were observed. Results: Compared with model group,the Fuzilizhong decoctions at high--dose,medium--dose and low-dose could reduce histological changes,the level of IL-6 and IL-8. The expressive levels of ICAM - 1 in colonic mueosa was reduced at the same time. The detection of indicators show that the effect of high dose group was better than that of SASP group, the difference has statistical significance. Conclusion : These results indicated that Fuzi Lizhong decoctions has effects on ulcerative colitis. Its mechanism is associated with decreased the content of Interleukin -6, interleukin -8 and the expressive levels of ICAM-1.
