白介素17E在过敏性哮喘患者嗜酸粒细胞表达的研究 被引量:1

摘要 目的 :评价白介素17E(IL-17E)对过敏性哮喘患者嗜酸粒细胞的作用。方法 :以流式细胞检测的方法检测14例过敏性哮喘患者、15例过敏性非哮喘患者、14名无过敏正常人嗜酸粒细胞IL-17E及受体的表达,以酶联免疫吸附法(ELISA)测定血浆IL-17E的水平,并比较了不同组别间的差异。结果:过敏性哮喘患者血浆IL-17E水平[145(64~290)ng/L]显著高于无过敏正常人群[21(0~116)ng/L](P=0.012)。血浆IL-17E水平与第1秒用力呼气容积(FEV_1)占预计值百分比(FEV_1%)呈显著负相关(r=-0.443,P=0.003)。嗜酸粒细胞表面IL-17E特异性受体IL-17R B的表达在过敏性哮喘患者中[43.08%(33.96%~59.98%)]显著高于过敏性非哮喘患者[11.98%(6.33%~27.11%)](P=0.002)和无过敏正常人群[17.70%(10.97%~38.9%)](P=0.01),IL-17RA的表达也在过敏性哮喘患者中[97.94%(94.25%~98.74%)]显著高于过敏性非哮喘者[86.60%(27.48%~97.64%)](P=0.036)和无过敏正常人群[93.24%(42.38%~97.36%)](P=0.027)。嗜酸粒细胞表面IL-17RB的表达与IL-17RA的表达相关(r=0.809,P=0.001)。过敏性哮喘患者血浆IL-17E水平为145(64~290)ng/L,显著高于无过敏正常人群[21(0~116)ng/L](P=0.012),但与过敏性非哮喘者[80(42~156)ng/L]比较无显著差异(P=0.275)。结论:过敏性哮喘患者血浆IL-17E水平、嗜酸粒细胞表达其受体IL-17RA和IL-17RB增高,提示IL-17E在过敏性炎症中嗜酸粒细胞的活化中发挥作用。 Objective To elucidate the effect of interleukin-17E(IL-17E) on eosinophils in allergic asthmatic patients.Methods IL-17 E, IL-17 RA and IL-17 RB expressions on eosinophils in normal subjects, atopic non-asthmatic and allergic asthmatic patients were detected by flow cytometry, and plasma level of IL-17 E was measured by enzyme linked immunosorbent assay(ELISA). The results of these measurements were analyzed and compared. Results Plasma level of IL-17 E increased significantly in allergic asthmatics when compared with normal controls [145(64-290) ng/L vs 21(0-116)ng/L,(P =0.012)], but no significant difference was found between allergic asthmatics and atopic non-asthmatics [80(42-156) ng/L,(P=0.275)]. Plasma IL-17 E level was negatively correlated with percentage of first minute forced expiratory volume to predicted value(FEV_1%)(r=-0.443, P=0.003). The expression of IL-17 RB on eosinophils was significantly higher in allergic asthmatics [43.08%(33.96%-59.98%)] than that in atopic non-asthmatics [11.98%(6.33%-27.11%)](P=0.002)and normal controls [17.70%(10.97%-38.9%)](P=0.01). The expression of IL-17 RA was also significantly higher in allergic asthmatics [97.94%(94.25%-98.74%)] than that in atopic non-asthmatics [86.60%(27.48%-97.64%)](P=0.036) and normal controls [93.24%(42.38%-97.36%)](P=0.027). The expression of IL-17 RB was positively correlated with expression of IL17RA(r=0.809, P =0.001). Conclusions Plasma level of IL-17 E and expressions of IL-17 RA and IL-17 RB on eosinophils are increased in patients with allergic asthma, and it is suggested that IL-17 E has some effects on activation of eosinophils in allergic inflammation.
