摘要
研究构建聚乙二醇(PEG)/硫酸铵[(NH_4)_2SO_4]双水相体系对乙醇脱氢酶(ADH)进行提取。探讨了PEG分子量、PEG浓度、(NH_4)_2SO_4浓度、粗酶添加量对ADH纯化倍数的影响。结果表明,在(NH_4)_2SO_4浓度为18%、PEG800浓度为22%、粗酶添加量为5%所组成的双水相体系下,ADH的纯化倍数可达3.815。
This study build polyethylene glycol(PEG)/ammonium sulfate [(NH4)2SO4] two water phase system extraction on ADH.The influences of PEG molecular weight,PEG concentration,(NH4)2SO4 concentration and crude enzyme additives on purification multiples were further investigated.The results showed that the purification multiples of ADH was 3.815 when the aqueous two-phase system were 18 %(NH4)2SO4,22.0 %PEG800 and 5 % crude enzyme additives.
