槲皮素对K562细胞增殖和凋亡的影响 被引量:2

摘要 目的探讨槲皮素(quercetin)对人慢性髓系白血病细胞K562的作用及其机制。方法培养K562细胞,分别用不同浓度(0,10,20μg/mL)的槲皮素处理。细胞培养48h后,利用MTT检测细胞增殖;流式检测K562细胞凋亡;RT-PCR检测细胞Bax和Bcl-2基因表达水平;检测细胞JAK2、STAT3、p-JAK2、p-STAT3、Bax和BCL-2蛋白浓度的变化。结果槲皮素诱导K562细胞增殖抑制和凋亡呈浓度依赖性,上调Bax表达,下调pJAK2、p-STAT3、Bcl-2表达,对JAK2和STAT3表达无明显影响。结论槲皮素能诱导K562细胞增殖抑制和凋亡,其机制可能是通过抑制JAK2/STAT3信号传导通路实现的。 Objective To explore the effect and mechanisms of the quercetin on the human leukemia K562 cells.Method The K562 cell was cultured in the DMEM.MTT method was used to evaluate the proli,feration effect of K562 and the apoptosis of K562 cells were evaluated by flow cytometric analysis.The mRNA level of Bax and Bcl-2were detected by RT-PCR.The expression level of JAK2,STAT3,p-JAK2,p-STAT3,Bax and Bcl-2protein were detected by Western blotting(WB).Result The result showed that quercetin treatment can significantly attenuate the proliferartion rate of K562 cell,the expression of p-JAK2,p-STAT3 and Bcl-2and increase the apoptosis rate of K562 cell,the expression of Bax.Therefore there were no influence in the expression JAK2 and STAT3.Conclusion Quercetin treatment could induce the growth inhibition and apoptosis of K562 cell and the effect may be achieved through suppressing JAK2/STAT3 signal transduction pathway activation.
