摘要
ω-6脂肪酸脱氢酶(FAD2)是亚油酸合成的关键酶,催化油酸脱氢形成亚油酸。以滇牡丹种子为材料,研究根据转录组数据设计的特异引物,采用RT-PCR方法,克隆滇牡丹ω-6脂肪酸脱氢酶(FAD2)并分析其功能。获得一个ω-6脂肪酸脱氢酶基因,将其命名为Pd FAD2。结果表明,该基因与芍药的同源性为98.7%。经氨基酸序列比对,推断滇牡丹FAD2具有植物FAD2特有的3个组氨酸区,包含完整的c DNA开放阅读框,由1 155bp组成,编码384个氨基酸残基。半定量PCR的结果显示,滇牡丹FAD2基因在花、茎、叶、种子中均有表达,在根中只有微弱表达。研究结果为研究滇牡丹不饱和脂肪酸代谢奠定基础,也为油用滇牡丹育种提供理论依据。
Omega-6 fatty acid desaturase (FAD2) is a critical enzyme in linoleic acid biosynthesis, and it converts oleic acid to linoleic acid by desaturation. In this study, an omega-6 fatty acid desaturase was obtained from Paeonia delavayi, which shared 98. 7% identities with that of Peaonia lactiflora, and named PdFAD2. Homology analysis showed that deduced PdFAD2 protein has three conserved histidine motifs. Expression profiling analysis revealed that PdFAD2 was expressed strongly in flower, stem, leaf and seed but slightly expressed in root. This result will provide basic information for unsaturated fatty acid biosynthesis research and molecular breeding in Paeonia delavayi.
