摘要
目的探讨呋虫胺原药对SD大鼠的免疫毒性,为呋虫胺的安全性评价提供科学依据。方法选择健康清洁级SD大鼠80只,按体重随机分为4组,每组20只,雌雄各半,设低、中、高3个剂量组,分别喂饲153.13 mg/kg、612.50 mg/kg和2 450.00 mg/kg的呋虫胺原药饲料。另设1个空白对照组,用清洁级普通标准饲料喂养。连续染毒90 d。观察4组大鼠脾脏、胸腺的脏体比以及常规组织病理学变化。结果与对照组比较,中、高剂量组大鼠脾脏、胸腺的脏体比降低,差异均有统计学意义(P<0.05),但低剂量组大鼠脾脏、胸腺的脏体比变化不明显(P>0.05);组织病理学检查发现,低倍镜下可见中、高剂量组大鼠脾脏白髓及胸腺皮质萎缩明显,高倍镜下均可见很多已陷入凋亡的淋巴细胞。结论呋虫胺可以引起大鼠免疫损伤,免疫细胞凋亡可能发挥着重要作用。
Objective To explore the immunotoxicity of dinotefuran in SD rats,which can provide a scientific basis for the safety evaluation of Dinotefuran. Methods The study was divided into low,medium and high dose groups,each dose group was fed 153. 13 mg / kg,612. 50 mg / kg and 2 450. 00 mg / kg of dinotefuran original medicated forage. In addition,a control group was set up,and fed with the clean- level common standard diet. All animals were exposed to the medicine for 90 days. Observe the change of dirty body ratio,histopathology of spleen and thymus. Results Compared with the control group,the dirty body ratio of spleen,thymus of rats is in middle,high dose group is lower,the differences are statistically significant( P〈0. 05),but the change of rats in low dose group is not obvious( P〉0. 05). Histopathological examination revealed the white pulp of spleen and cortical of thymus in the high dose group rats have obvious atrophy at low magnification microscope,it was seen a lot of lymphocytes which has fallen into apoptosis at high magnification. Conclusion Dinotefuran could induce immune damage,and immune cell apoptosis may play an important role.
