腺病毒介导ACE2基因对鼻咽癌细胞株CNZ-2 Z增殖能力的影响

Effect of adenovirus mediated ACE2 on proliferation of nasopharyngeal carcinoma cell line CNZ-2Z

目的：观察腺病毒介导血管紧张素转换酶-2（ACE2）基因对鼻咽癌细胞株CNE-2Z增殖能力的影响。方法构建稳定高表达ACE2的鼻咽癌细胞株CNE-2Z,免疫印迹法检测转染前后ACE2蛋白表达的变化,采用MTT法及克隆形成实验检测转染前后细胞增殖能力、体外成瘤能力的改变。结果腺病毒介导的ACE2表达质粒能有效上调CNE-2Z细胞中ACE2蛋白的表达。上调CNE-2Z细胞中ACE2的表达后,CNE-2Z细胞的增殖能力及体外成瘤能力均明显受到抑制（P〈0．05）。结论腺病毒介导ACE2基因能明显抑制鼻咽细胞CNE-2Z在体外的增殖及成瘤能力。

Objective To investigate the effect of adenovirus mediated ACE2 on proliferation of nasopharyngeal carcinoma cell line CNZ-2Z. Methods ACE2 expression plasmid was designed and was transfected into nasopharyngeal carcinoma cell line CNZ-2Z. The expression of ACE2 protein in CNZ-2Z was detected by western blot. The proliferation and tumorigenicity of cancer cells were tested by MTT and clone formation experiment. Results ACE2 protein ex-pression was upregulated after ACE2 expression plasmid transfection. The proliferation and clone formation ability of CNZ-2 Z cells transfected with ACE2 expression plasmid were significantly suppressed compared with control cells （P〈0. 05）. Conclusions Upregulation of ACE2 can significantly inhibit the proliferation and clone formation abil-ity in CNZ-2 Z cells.
